PROT (Sodium-dependent proline transporter), also known as Solute carrier family 6 member 7, is a 636 amino acid multi-pass membrane protein that is a GAT1 family member specifically expressed in regions of the brain. PROT terminates the action of proline by its high affinity sodium/chloride-dependent reuptake into pre-synaptic terminals. Enriched in glutamatergic synaptic terminals, it is likely that PROT plays an important role in excitatory events of neurotransmission. PROT-mediated proline uptake is inhibited by compounds such as benztropine, LP-403812 and Des-Tyr-Leu-enkephalin (GGFL). These inhibitors of proline uptake may lead to the development of therapeutic agents for certain neurologic disorders.

Noxa Antibody: Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes including p53R2, p53AIP1, and PUMA. A new p53 target gene, Noxa, was recently identified, which encodes a protein belonging to the subfamily of BH3-only proapoptic proteins. Noxa and PUMA are both transcriptional targets of p53 and BH3-only proteins. X-ray irradiation increased p53-dependent Noxa mRNA and protein levels. Noxa, when ectopically expressed, interacted with anti-apoptotic Bcl-2 family members, resulting in the activation of caspase-9. Noxa, like PUMA, localized to mitochondria and induces apoptosis in response to p53. Noxa and PUMA may represent direct mediators of p53-induced apoptosis. Increased levels of p53 and its target gene Noxa was found in the impaired tumor development.

Three isoforms of p53AIP are known to exist; this antibody will detect all three.

Bcl-rambo Antibody: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. Members of the Bcl-2 family are known to be critical regulators of this process. These proteins are characterized by the presence of several conserved motifs termed Bcl-2 homology (BH) domains. A novel, widely expressed member termed Bcl-rambo was recently identified. This protein is localized to mitochondria in mammalian cells and its overexpression induces apoptosis which could be blocked by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2. Bcl-rambo shows overall homology to the anti-apoptotic members containing BH motifs, but unlike Bcl-2, the C-terminal membrane anchor of Bcl-rambo is preceded by a unique 250 amino acid insertion. This region by itself can induce apoptosis more efficiently than the Bcl-2 homology regions, suggesting that Bcl-rambo may be important other pro-apoptotic pathways.
