Anti-CD45.2 Mouse Monoclonal Antibody (PerCP (Peridinin-Chlorophyll Protein Complex)-Cy5.5®) [clone: 104]

Supplier: BIOGEMS INTERNATIONAL INC.

The 104 monoclonal antibody specifically reacts with the mouse CD45 molecule, the Leukocyte Common Antigen (LCA) which occurs in the alloantigen CD45.2-expressing mouse strains (C57BL/6, CBA, 129, A, AKR, C58, DBA/1, DBA/2, BALB/c, and C3H/He). The 104 monoclonal antibody does not react with the leukocytes of the CD45.1-expressing mouse strains (DA, SJL/J, RIII, and STS/A). The CD45 molecule is a member of the Protein Tyrosine Phosphatase (PTP) family, because its intracellular region contains two PTP domains. The extracellular region’s variability is caused by different levels of glycosylation, and the splicing of the 4, 5, and 6 exons. The isoforms found in the mouse strains depend on the activation state, maturation stage and cell type, and are very important in B and T lymphocytes antigen receptor signal transduction. The 104 antibody inhibits some of the B lymphocytes responses, reduces the serum IgG levels, and influences the autoimmune renal pathology.

Mouse Recombinant sCD40 (from E. coli)

Supplier: Peprotech

CD40, a member of the TNF receptor superfamily, is a cell surface protein expressed on B cells, dendritic cells, monocytes, thymic epithelial cells and, at low levels, on T cells. Signaling though CD40 plays an important role in the proliferation and differentiation of B cells, and is critical for immunoglobulin (Ig) class switching. The membrane-anchored CD40-Ligand is expressed almost exclusively on activated CD4+ T lymphocytes. Failure to express CD40L leads to “immunodeficiency with hyper-IgM”, a disease characterized by failure to produce IgG, IgA and IgE. The human CD40L gene codes for a 261 amino acid type II transmembrane protein, which contains a 22 amino acid cytoplasmic domain, a 24 amino acid transmembrane domain, and a 215 amino acid extracellular domain. The soluble form of CD40L is an 18 kDa protein comprising the entire TNF homologous region of CD40L and is generated in vivo by an intracellular proteolytic processing of the full length CD40L. Recombinant Murine sCD40 Ligand is a soluble 16.4 kDa protein containing 149 amino acid residues comprising the receptor binding TNF-like domain of CD40L.

Anti-VhH Rabbit Polyclonal Antibody

Anti-VhH Rabbit Polyclonal Antibody

Supplier: Rockland Immunochemical

Rabbit Anti-VhH antibody can be used to detect single domain VhH antibodies. The single-domain antibody (sdAb) is a small (12 - 14 kDa) antibody fragment that consists of a monomeric variable domain derived from the heavy chain, also called a VhH antibody. These heavy chain only Fab-like domains have activity that is similar to a whole antibody, and they are able to bind to a specific antigens. SdAb's are derived from camelid species that include llamas, alpacas and camels. Camelids produce both classical (containing heavy and light chain fragments) and non-classical antibody structures (containing only a heavy chain). VhH antibodies are the smallest functional antigen-binding fragment that occurs in nature and these are now being used in biotechnology as a novel antibody scaffold. The small size of the VhH single domain antibody makes it very attractive for use in diagnostic imaging and they have potential for therapeutic activity.

Anti-CD3D Monoclonal Antibody [clone: UCHT1] (APC (Allophycocyanin))

Anti-CD3D Monoclonal Antibody [clone: UCHT1] (APC (Allophycocyanin))

Supplier: Tonbo Biosciences

The UCHT1 antibody is specific for human CD3e, also known as CD3 epsilon, a 20 kDa subunit of the T cell receptor complex, along with CD3 gamma and CD3 delta. These integral membrane protein chains assemble with additional chains of the T cell receptor (TCR), as well as CD3 zeta chain, to form the T cell receptor – CD3 complex. Together with co-receptors CD4 or CD8, the complex serves to recognize antigens bound to MHC molecules on antigen-presenting cells. These interactions promote T cell receptor signaling (T cell activation), inducing cell proliferation, differentiation, production of cytokines or activation-induced cell death. CD3 is differentially expressed during thymocyte-to-T cell development and on all mature T cells.

Anti-CD28 Mouse Monoclonal Antibody (PE (Phycoerythrin)-Cy7®) [clone: CD28.2]

Anti-CD28 Mouse Monoclonal Antibody (PE (Phycoerythrin)-Cy7®) [clone: CD28.2]

Supplier: Tonbo Biosciences

The CD28.2 antibody reacts with human CD28, a 44 kDa type I surface glycoprotein which acts as a co-stimulatory receptor in support of the T cell receptor (TCR). CD28 exists as a homodimer with specificity for two known ligands, known as B7-1 (CD80) and B7-2 (CD86), which are expressed on activated B cells and antigen-presenting cells. These ligands trigger CD28 signaling in concert with TCR activation to drive T cell proliferation, induce high-level expression of IL-2, impart resistance to apoptosis, and enhance T cell cytotoxicity. The interaction / co-stimulatory signaling between the B7 ligands and CD28 provides crucial communication between T cells and B cells or APCs to coordinate the adaptive immune response. Other members of the CD28 family of receptors include CTLA-4 (CD152), PD-1 (CD279), ICOS and BTLA.

Anti-AKT3 Rabbit Polyclonal Antibody (Biotin)

Supplier: Rockland Immunochemical

AKT is a component of the PI-3 kinase pathway and is activated by phosphorylation at Ser 473 and Thr 308. AKT is a cytoplasmic protein also known as AKT1, Protein Kinase B (PKB) and rac (related to A and C kinases). AKT is a key regulator of many signal transduction pathways. AKT Exhibits tight control over cell proliferation and cell viability. Overexpression or inappropriate activation of AKT is noted in many types of cancer. AKT mediates many of the downstream events of PI 3-kinase (a lipid kinase activated by growth factors, cytokines and insulin). PI 3-kinase recruits AKT to the membrane, where it is activated by PDK1 phosphorylation. Once phosphorylated, AKT dissociates from the membrane and phosphorylates targets in the cytoplasm and the cell nucleus. AKT has two main roles: (i) inhibition of apoptosis; (ii) promotion of proliferation. Anti-AKT Antibody is ideal for investigators involved in Cell Signaling, Cancer, Neuroscience, Signal Transduction research.

Anti-TBC1D10C Rabbit Polyclonal Antibody

Anti-TBC1D10C Rabbit Polyclonal Antibody

Supplier: Rockland Immunochemical

Antigen binding by the T-cell receptor (TCR) is one of the critical first steps in the immune response, triggering a cascade of signaling pathways that ultimately lead to T-cell activation. Screening a yeast two-hybrid screen of a human T-cell cDNA library with calcineurin, a protein phosphatase involved in multiple signaling pathways including T-cell activation, resulted in the identification of Carabin, a member of the TBC1 domain family of proteins, as a calcineurin-binding protein. Unlike other members of the TBC1 domain protein family which are thought to have a role in regulating cell growth and differentiation, further experiments demonstrated that Carabin is part of a negative regulatory loop for the intracellular TCR signaling pathway as well as an inhibitor of the Ras signaling pathway, suggesting that Carabin may also mediate crosstalk between calcineurin and Ras. Carabin antibody does not recognize TBC1D10A or TBC1D10B. Carabin is known to exist in multiple isoforms.

Anti-HSP25 Mouse Monoclonal Antibody [clone: G3.1]

Anti-HSP25 Mouse Monoclonal Antibody [clone: G3.1]

Supplier: Enzo Life Sciences

Hsp27 is one of the most common members of the highly conserved and ubiquitously expressed family of small heat shock proteins (sHsp), which also includes alphaB-crystallin. It is characterized by a conserved C-terminal alpha-crystallin domain consisting of two anti-parallel beta-sheets that promote oligomer formation required for its primary chaperone function as inhibitor of irreversible protein aggregation. Hsp27 oligomerization is modulated by post-translational phosphorylation of Hsp27 at three serine residues, Ser15, Ser78, and Ser82, by a variety of protein kinases including MAPKAPK-3, PKAc-alpha, p70 S6K, PKD I, and PKC-delta. Hsp27 has been shown to inhibit actin polymerization by binding of unphosphorylated Hsp27 monomers to actin intermediate filaments. Anti-apoptotic functions of Hsp27 have also been identified through interactions with DAXX7, activation of Akt, and inhibition of apoptosome formation. Evidence suggests altered expression of Hsp27 is implicated in the pathogenesis of breast, ovarian, and prostate cancer.

Human Recombinant IL-12 (from CHO cells)

Supplier: Adipogen

Interleukin-12 (IL-12), also known as Natural Killer Cell Stimulatory Factor (NKSF) or Cytotoxic Lymphocyte Maturation Factor (CLMF), is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. IL-12 is produced by macrophages and B lymphocytes and has been shown to have multiple effects on T cells and Natural Killer (NK) cells. Some of these IL-12 activities include the induction of IFN-gamma and TNF in resting and activated T and NK cells; the enhancement of cytotoxic activity of resting NK and T cells, the stimulation of resting T cell proliferation in the presence of a comitogen; and the enhancement of NK cell proliferation. Current evidence indicates that IL-12 is a key mediator of cellular immunity and induces the differentiation of Th1 cells from precursor T helper cells. Based on its activities, it has been suggested that IL-12 may have therapeutic potential as a vaccine adjuvant that promotes cellular immunity and as an antitumor and anti viral agent.

Anti-CD8a Rat Monoclonal Antibody (PE (Phycoerythrin)) [clone: 53-6.7]

Supplier: BIOGEMS INTERNATIONAL INC.

The 53-6.7 monoclonal antibody specifically reacts with Ly-2, the 38 kDa alpha chain, and with Lyt-2, the 34 kDa alpha’ chain, of the mouse CD8 antigen. The alpha’ chain is the truncated form of alpha chain, encoded by the same CD8a gene. In CD8a, the alpha and alpha’ chains form heterodimers with CD8b (the beta chains) or homodimers (alpha-alpha), which occur as receptors on the surface of the majority of thymocytes. A subpopulation of mature T lymphocytes expresses the CD8 alpha beta (alpha beta TCR T cells), and a subpopulation of intestinal intraepithelial lymphocytes and dendritic cells express CD8a without CD8b.CD8 interacts with the mouse major histocompatibility complex class I (MHC class I) molecules on antigen-presenting cells or epithelial cells. Its function seems to be to attenuate the CD8-mediated signal for the stimulation of intrathymic T-cell maturation. The 53-6.7 antibody is useful for depleting CD8+ peripheral T lymphocytes. It cross reacts with the alpha- and alpha’-like polypeptides on some thymic and peripheral lymphocytes.

Human Recombinant CD137 (from E. coli)

Supplier: Prosci

Human CD137 (4-1BB) is a costimulatory molecule of the tumor necrosis factor (TNF) receptor superfamily. The glycoprotein 4-1BB is expressed mainly on activated CD4+ and CD8+ T cells and binds to a high-affinity ligand (4-1BBL) expressed on several antigen-presenting cells such as macrophages and activated B cells. Upon ligand binding, 4-1BB is associated with the tumor receptor-associated factors (TRAF), the adaptor protein and mediates downstream signaling events including the activation of NF-kappaB and cytokine production. 4-1BB signaling either by binding to 4-1BBL or by antibody ligation delivers signals for T cell activation and growth as well as monocyte proliferation and B cell survival, and plays a important role in the amplification of T cell-mediated immune responses. In contrast, it can also enhance activation-induced T cell apoptosis when triggered by engagement of the TCR/CD3 complex. In addition, the 4-1BB/4-1BBL costimulatory pathway has been shown to augment secondary CTL responses to several viruses and increase antitumor immunity. 4-1BB is therefore a promising candidate for immunotherapy of human cancer.
