Recombinant human PYK2 (360-690) was expressed by baculovirus in Sf9 cells using an N-terminal His tag. PYK2 (also known as FAK2/RAFTK) is a member of the focal adhesion PTK family. PYK2/FAK2 can be activated by a variety of extracellular signals that elevate intracellular calcium concentration and by stress signals (1). Unlike FAK, which is widely expressed in various tissues and links transmembrane integrin receptors to intracellular pathways, PYK2/FAK2 is expressed mainly in the central nervous system and in cells derived from hematopoietic lineages. In osteoclasts, although FAK is expressed, PYK2/FAK2 appears to be the predominant mediator of integrin alpha(v)beta3 signaling events that influence osteoclast physiology and pathology (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: PYK2, 10ug (Human, recombinant; amino acids 360-690). MW: ~39kDa. Substrate: Poly (4:1 Glu, Tyr) peptide. Other: Reaction Buffer, DTT, MnCl2.
