Recombinant human TGF beta R1 (ALK5) (80-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. TGF beta R1 or transforming growth factor beta-receptor 1 is a member of the TGF beta receptor subfamily and is a ser/thr protein kinase that forms a heteromeric complex with type II TGF-beta receptors when bound to TGF-beta, transducing the TGF beta signal from the cell surface to the cytoplasm. Mutations in the TGF beta R1 gene have been associated with Marfan syndrome, Loeys-Deitz Aortic Aneurysm Syndrome and the development of various types of tumors (1). TGF beta R1-dependent signaling is required for angiogenesis but not for the development of hematopoietic progenitor cells and functional hematopoiesis (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
