About this item

Recombinant full-length human p70S6Kb was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. p70S6Kb is a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases and is activated by mitogenic stimuli.

  • Easily Screen and Profile p70S6Kb Kinase Inhibitors
  • Includes kinase, substrate and reaction buffer
  • Use with ADP-Glo™ Assay for bioluminescent detection of kinase activity

Recombinant full-length human p70S6Kb was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. p70S6Kb is a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases and is activated by mitogenic stimuli, including growth factors, cytokines and phorbol esters. p70S6Kb contains 2 nonidentical kinase catalytic domains and phosphorylates the S6 ribosomal protein and eukaryotic translation initiation factor 4B. Phosphorylation of S6 leads to an increase in protein synthesis and cell proliferation. PI3 kinase pathway and mTOR are involved in the activation of p70S6Kb, but other pathways can also activate this target protein. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: p70S6Kb, 10ug (Human, recombinant full-length). MW: ~85kDa. Substrate: RSK substrate (KRRRLSSLRA); derived from human 40S ribosomal protein S6 (amino acids 230-239). Other: Kinase Assay Buffer I, DTT. p70S6Kb NCBI Database Entry: www.ncbi.nlm.nih.gov/gene/6199/. Visit www.promega.com/kinase/ to see all Kinase Enzyme Systems.
