CK1 alpha 1 is a member of the CK1 family of serine/threonine protein kinases, which play an important role in diverse cell processes. CK1 can regulate Smo cell surface accumulation and activity in response to hedgehog. CK1 phosphorylates Smo at several sites, and phosphorylation-deficient forms of Smo fail to accumulate on the cell surface and are unable to transduce the hedgehog signal (1). CK1 alpha 1 dynamically associates with the CBM complex on T cell receptor engagement to participate in cytokine production and lymphocyte proliferation. CK1 alpha 1 can form complexes with MDM2, which then regulates the stability of p53 and E2F-1 transcription factors (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
