Vasopressin, also known as arginine vasopressin (AVP) or antidiuretic hormone (ADH), is a posterior pituitary hormone that is synthesised in the hypothalamus. Vasopressin is synthesised as a precursor protein that consists of arginine vasopressin and two associated proteins, neurophysin 2 and the glycopeptide copeptin. Vasopressin, together with its carrier protein neurophysin II, is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis, where it is either stored or secreted into the bloodstream. Vasopressin acts as a growth factor by enhancing pH regulation through acid-base transport systems. It has a direct antidiuretic action on the kidney and also causes vasoconstriction of the peripheral vessels. Vasopressin can also contract smooth muscle during parturition and lactation. It also plays a role in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions. Mutations in the vasopressin precursor cause autosomal dominant neurohypophyseal diabetes insipidus (ADNDI), which is characterised by persistant thirst, polydipsia and polyuria.

SerpinB2 is a serine proteinase inhibitor of the ovalbumin like B clade of serpins. It was first discovered in the placenta, and given the name PAI-2 because of the ability to inhibit urokinase plasminogen activator (u-PA) at low micromolar efficiency. SerpinB2 also inhibits tissue plasminogen activator (t-PA), but with micromolar efficiency, and PAI-1 is much more efficient that SerpinB2 on both forms of plasminogen activator. The structure of PAI-2 is not terribly similar to PAI-1, however, which is an E clade serpin. SerpinB2 is made by many cell types, and is found intracellularly as an unglycosylated kDa protein, and secreted as a 60 kDa protein. SerpinB2 is found in saliva, secreted by gingival fibroblasts, and in the skin. SerpinB2 levels are elevated in serum during pregnancy, and in leukemia, breast cancer and ovarian cancer, although it was lowered in some cancers.A shorter SerpinB2 isoform of 382 amino acids, has been reported, with a predicted mass of 43.1 kDa and a pI of 5.69. The shorter form has a deletion just after the start of the mature protein, but it is unclear what the relative production and distribution is for the shorter sequence.

The eight members of the recently identified Suppressor of Cytokines Signaling (SOCS) family are SOCS1, SOCS2, SOCS3, SOCS4, SOCS5, SOCS6, SOCS7, and CIS. Structurally the SOCS proteins are composed of an N- terminal region of variable length and amino acid composition, a central SH2 domain, and a C-terminal motif called the SOCS box. The SOCS proteins appear to form part of a classical negative feedback loop that regulates cytokine signal transduction. Transcription of each of the SOCS genes occurs rapidly in vitro and in vivo in response to cytokines, and once produced, the various members of the SOCS family appear to inhibit signaling in different ways. During Th1 differentiation a reduction in the association of Jak1 with the IL4 Receptor correlated with the appearance of SOCS5. SOCS5 protein was preferentially expressed in committed Th1 cells and interacted with the cytoplasmic region of the IL4 Receptor alpha chain irrespective of receptor tyrosine phosphorylation. This unconventional interaction of SOCS5 protein with IL4 Receptor resulted in the inhibition of IL4-mediated signal transducer and activator of transcription-6 activation. T cells from transgenic mice constitutively expressing SOCS5 exhibited a significant reduction of IL4-mediated Th2 development. Therefore, the induced SOCS5 protein in Th1 differentiation environment may play an important role by regulating Th1 and Th2 balance.
