Human EN-RAGE belongs to the S100/calgranulin superfamily. There are at least 21 different S100 proteins and the protein is 100% soluble in ammonium sulfate at neutral pH. S100 proteins play a role in regulation of protein phosphorylation, transcription factors, the dynamics of cytoskeleton constituents, enzyme activities, cell growth and differentiation, and the inflammatory response.Human EN-RAGE is characterized by two EF-hand calcium-binding motifs, zinc- and copper-binding protein.S100A12 is a disulfide-linked homodimer and the interface between the two subunits is composed mostly of hydrophobic residues.Its proinflammatory activity involves recruitment of leukocytes, promotion of cytokine and chemokine production, and regulation of leukocyte adhesion and migration. EN-RAGE acts as an alarmin or a danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to receptor for advanced glycation endproducts (AGER). Binding to AGER activates the MAP-kinase and NF-kappa-B signaling pathways leading to production of proinflammatory cytokines and up-regulation of cell adhesion molecules ICAM1 and VCAM1. It also acts as a monocyte and mast cell chemoattractant. Moreover, it can stimulate mast cell degranulation and activation which generates chemokines, histamine and cytokines inducing further leukocyte recruitment to the sites of inflammation. It can inhibit the activity of matrix metalloproteinases; MMP2, MMP3 and MMP9 by chelating Zn2+from their active sites.

The procedure for determining dietary fibre exposes the sample to a series of enzymatic digestions that simulate the real digestive process which takes place in the human or animal digestive tract, calculating the undigested residue remaining at the end of the analysis. Generally dietary fibre analysis is carried out on foods intended for human consumption whereas raw fibre analysis is carried out on animal feeds or on raw materials of vegetable origin, eg. cereals. The GDE performs enzymatic digestion, a delicate phase where samples are immersed in a thermostatic water bath and stirred. Continuous and constant sample mixing is necessary in order to prevent the sample from overheating. The unit consists of an immersion thermostat, a transparent polycarbonate tank and a VELP 6-place magnetic stirrer to ensure excellent thermoregulation and precision. The use of deionised water is recommended as it eliminates any possible cause of limescale.

Fibroblast growth factor 16 (FGF-16) is a heparin-binding member of the FGF family. FGFs possess broad mitogenic and cell survival activities and are expressed during embryonic development. tumour Necrosis Factor- alpha (TNF- alpha) is secreted by macrophages, monocytes, neutrophils, T-cells, and NK-cells following stimulation by bacterial LPS. Cells expressing CD4 secrete TNF- alpha while cells that express CD8 secrete little or no TNF- alpha. Synthesis of TNF- alpha can be induced by many different stimuli including interferons, IL2, and GM-CSF. The clinical use of the potent anti-tumour activity of TNF- alpha has been limited by the proinflammatory side effects such as fever, dose-limiting hypotension, hepatotoxicity, intravascular thrombosis, and hemorrhage. Designing clinically applicable TNF- alpha mutants with low systemic toxicity has been of intense pharmacological interest. Human TNF- alpha that binds to murine TNF-R55 but not murine TNF-R7, exhibits retained anti-tumour activity and reduced systemic toxicity in mice compared with murine TNF- alpha, which binds to both murine TNF receptors. Based on these results, many TNF- alpha mutants that selectively bind to TNF-R55 have been designed. These mutants displayed cytotoxic activities on tumour cell lines in vitro and have exhibited lower systemic toxicity in vivo. Recombinant Human TNF- alpha High Active Mutant differs from the wild-type by amino acid subsitution of amino acids 1-7 with Arg8, Lys9, Arg10 and Phe157. This mutant form has been shown to have increased activity with less inflammatory side effects in vivo.
