Mouse Recombinant IL-10
Supplier: STEMCELL Technologies
Interleukin 10 (IL-10) is the founding member of the IL-10 family of class II cytokines. All of the IL-10 cytokine family members have a four-helix bundle consisting of α-helical folds. Upon binding to its receptor, IL-10 activates signaling through JAK1 and STAT3. It is produced by dendritic cells, macrophages, and CD4+ T regulatory cells, as well as mast cells, NK cells, neutrophils, and regulatory B cells, under specific stimulating conditions (Saraiva and O'Garra). IL-10 can inhibit the activation of certain immune cells while it promotes the function of B cells, and facilitate healing process. Specifically, this cytokine is important for the function of T regulatory cells as it is a potent suppressor of effector T cell proliferation and cytokine production. Also, IL-10 produced by a subset of macrophages inhibits activation and production of pro-inflammatory cytokines by neighboring macrophages, thus allowing a level of self-regulation. IL-10 enhances B cell proliferation, immunoglobulin secretion, and class II MHC expression (Ouyang et al.).

Intrinsic factor (from porcine pyloric mucous lining), powder
Supplier: MP Biomedicals
Preparation Method: MP Intrinsic Factor is produced from the pylorus mucous membrane of the pig, of which only a small part is used. Only the part which, according to histological examinations has proved to have a high content of Intrinsic Factor, is used.
Applications: Intrinsic Factor is used in the treatment not only of classical pernicious anaemia, but also in the treatment of other macrocytic anaemias, where it replaces the Intrinsic Factor which may be lacking in the patient's own gastric juice. In this way Intrinsic Factor is recommended against anaemias that appear 6 to 12 months after serious gastric operations, and against anaemias that are secondary effects of the treatment with certain medicines, e.g. against epilepsy. However, the dietetic use in multi-vitamin preparations is of the greatest importance. In older people there has been observed a degeneration of the gastric mucous membrane, whose secretion of Intrinsic Factor is insufficient to ensure the resorption of the necessary quantity of B-12 from their diet. In order to delay this development it is useful to prescribe a small quantity of Intrinsic Factor with B-12 and other vitamins to these patients.
Product Description: Intrinsic factor is a glucoprotein extracted from the pylorus part of the gastric mucous membrane. Its presence is considered necessary for the absorption of Vitamin B-12.
Unit Definition: The activity is normally indicated in Coli units, the number of which are proportional to the binding power of the substance to Vitamin B-12.
Application Areas: Absorption of vitamin B12
Product Type: Proteins, Enzymes & Peptides
Presentation: Tan Powder
Foreign activity: Salmonella=Negative, E. coli=Negative
NOTES: Standard Plate Count <1000/g

Mouse recombinant IL36 alpha (from E. coli)
Supplier: ProSci Inc.
Mouse Interleukin-36 alpha(Il-36a)is a member of the IL-1 family.IL¬1 alpha,IL¬1 beta and IL-18 are potent inflammatory cytokines whose activities are dependent on heterodimeric receptors of the IL-1R superfamily, and which are regulated by soluble antagonists. Il36a is expressed in many cells, including monocytes, B cells, and T cells, immune system and fetal brains. Il36a is a cytokine that binds to and signals through the IL1RL2/IL-36R receptor which in turn activates NF-kappa-B and MAPK signaling pathways in target cells linked to a pro-inflammatory response. It is a part of the IL-36 signaling system that is thought to be present in epithelial barriers and to take part in local inflammatory response; similar to the IL-1 system with which it shares the coreceptor IL1RAP. It seems to be involved in skin inflammatory response by acting on keratinocytes, dendritic cells and indirectly on T cells to drive tissue infiltration, cell maturation and cell proliferation. It Induces the production of proinflammatory cytokines, including IL-12, Il-1 beta, IL-6, TNF-alpha and IL-23 in bone marrow-derived dendritic cells (BMDCs). Moreover, it is involved in dendritic cell maturation by stimulating the surface expression of CD80, CD86 and MHC class II and can induce the production of IFN-gamma, IL-4 and IL-17 by cultured CD4+ T cells and splenocytes. Il36a may play a role in proinflammatory effects in the lung: induces the expression of CXCL1 and CXCL2 in the lung, and the expression of TNF-alpha, IL-36c, IL-1A, IL-1B, CXCL1 and CXCL2 in isolated splenic CD11c+ alveolar macrophages. It may be involved in T cell maturation by stimulating the surface expression of CD40 and modestly CD80 and CD86 in splenic CD11c+ cells and CD4+ T cell proliferation.

Human Recombinant CD40 Ligand
Supplier: STEMCELL Technologies
CD40 ligand is a type II transmembrane glycoprotein that belongs to the tumor necrosis factor (TNF) superfamily (Quezada et al.). CD40 ligand forms a bioactive homotrimer that exists as both soluble and membrane-bound forms (Khandekar et al.). CD40 ligand is expressed on T cells, monocytes, basophils, eosinophils, platelets, dendritic cells, and endothelial cells. Its receptor, CD40, is expressed on B cells, dendritic cells, macrophages, monocytes, platelets, endothelial cells, and epithelial cells (van Kooten and Banchereau). Binding of CD40 ligand to CD40 stimulates B cell proliferation, immunoglobulin class switching, antibody secretion, and T cell-dependent humoral responses. Dysregulation of CD40 ligand contributes to immune deficiency in HIV and AIDS (Rickert et al.). CD40 ligand has also been linked to the pathology of atherosclerosis, atherothrombosis, and restenosis (Hassan et al.).

Human recombinant IL20 (fromE. coli)
Supplier: ProSci Inc.
Interleukin-20 (IL-20) is a member of the IL-10 family of regulatory cytokines that includes IL-10, IL-19, IL-20, IL-22, IL-24 and IL-26. Members of this family share partial homology in their amino acid sequences but they are dissimilar in their biological functions. IL-20 exhibits approximately 28% amino acid identity with IL-10 and 76% amino acid identity with mouse IL-20. There are two heterodimeric receptor complexes for IL-20. The first is composed of IL-20 R alpha and IL-20 R beta . The second is composed of IL-22 R and IL-20 R beta . Whereas the IL-22 R/IL-20 R beta complex is shared with IL-24, the IL-20 R alpha/IL-20 R beta complex is shared with both IL-19 and IL-24. IL-20 has been shown to initiate transduction cascades involving STAT3 and stimulates the induction of pro-inflammatory genes including TNF- alpha and MCP-1. Initial functional studies using transgenic mice suggest that IL-20 has the ability to regulate skin development. The over-expression of both human and mouse forms of IL-20 results in keratinocyte hyper-proliferation, abnormal epidermal differentiation, and neonatal lethality. In humans, IL-20 and its receptors are up-regulated in psoriatic skin, and polymorphisms in the IL-20 gene have been associated with plaque-type psoriasis. IL-20 may also have a role in hematopoiesis. It enhances the proliferation of multi-potential progenitors in vitro and increases their numbers and cell cycling status in IL-20 transgenic mice. IL-20 is also shown to suppress COX-2 and PGE2 and acts as an inhibitor of angiogenesis in model systems.

Human Recombinant Sclerostin, His Tag
Supplier: STEMCELL Technologies
Use sclerostin to alter bone remodeling homeostasis, where it inhibits bone formation in vivo and in vitro, likely through Wnt/β-catenin signaling cascades (Ellies et al.; Lin et al.). Sclerostin is a member of the cerberus/DAN family of glycoproteins whose members share a C-terminal cysteine-knot-like (CTCK) domain. Highly expressed in bone and cartilage, as well as in kidney, and liver tissues (Weivoda et al.), this osteoclast-derived BMP antagonist binds BMP6 and BMP7 with high affinity, and binds BMP2 and BMP4 with a lower affinity (Kusu et al.). Mutations in the SOST gene have been associated with sclerosteosis (Brunkow et al.), van Buchem disease (Staehling-Hampton et al.), and bone dysplasia disorders, characterized by increased bone density. This protein product contains a His-residue tag at the carboxyl end of the polypeptide chain. For consistency and reproducibility across your applications, sclerostin from STEMCELL comes lyophilised with ≥87% purity, and endotoxin levels are verified to be ≤1.0 EU/μg protein.

Human Recombinant 15-PGDH, His tag
Supplier: STEMCELL Technologies
Modulate prostaglandin metabolism with 15-hydroxyprostaglandin dehydrogenase (15-PGDH). 15-PGDH catalyses the reversible oxidation of the 15-hydroxyl group in prostaglandins, resulting in inactivated metabolites (Ensor and Tai), it can act on a variety of prostaglandins as substrates in a NAD+ dependent manner (Cho et al.). As prostaglandins can have a range of effects on cellular processes, 15-PGDH is of considerable importance in drug development. In vitro and in vivo studies suggest 15-PGDH has tumor suppressive activity and is downregulated in certain cancers (Na et al.).15-PGDH belongs to the short-chain dehydrogenases/reductases (SDR) family; its primary structure indicates 20% homology with other SDRs, with some conserved amino acid residues (Krook et al.). This protein contains a His-residue tag at the carboxyl end of the polypeptide chain. For consistency and reproducibility across your applications, apo M from STEMCELL comes lyophilised with ≥89% purity.

Human recombinant Fc gamma RIIa (from cells)
Supplier: ProSci Inc.
Receptors for the Fc region of IgG (Fc gamma R) are members of the Ig superfamily that function in the activation or inhibition of immune responses. Human Fc gamma Rs are divided into three classes designated Fc gamma RI (CD64), Fc gamma RII (CD32), and Fc gamma RIII (CD16), which generate multiple isoforms, are recognized. The activating type receptor either has or associates noncovalently with an accessory subunit that has an immunoreceptor tyrosinebased activation motif (ITAM) in its cytoplasmic domain. Fc gamma RI binds IgG with high affinity and functions during early immune responses, whereas Fc gamma RII and RIII are low affinity receptors that recognize IgG as aggregates surrounding multivalent antigens during late immune responses. Three genes for human Fc gamma RII (A, B, and C) and one for mouse (Fc gamma RIIB), encoding type I transmembrane proteins with ITAM motifs (Fc gamma RII A and C) or ITIM motifs (Fc gamma RIIB) in their cytoplasmic domains, have been identified. Human CD32, also known as Low affinity immunoglobulin gamma Fc region receptor II-a (IgG Fc receptor II-a), Fc gamma RII A or FCGR2A Protein, is expressed on cells of both myeloid and lymphoid lineages as well as on cells of non-hematopoietic origin. Associated with an ITAM-bearing adapter subunit, FcR gamma , CD32a (Fc gamma RII A) delivers an activating signal upon ligand binding, and results in the initiation of inflammatory responses including cytolysis, phagocytosis, degranulation, and cytokine production. The responses can be modulated by signals from the co-expressed inhibitory receptors such as Fc gamma RII B, and the strength of the signal is dependent on the ratio of expression of the activating and inhibitory receptors.

Human Recombinant VEGF-C
Supplier: STEMCELL Technologies
Vascular endothelial growth factor C (VEGF-C) is a member of the VEGF/platelet-derived growth factor (PDGF) family of proteins. VEGF-C is a potent angiogenic factor and promotes lymphangiogenesis, endothelial cell growth and survival, and can affect blood vessel permeability. VEGF-C is expressed in a range of tissues, but is not expressed in peripheral blood lymphocytes. VEGF-C forms a non-covalent, cell surface-associated, disulfide-linked homodimer that can bind and activate VEGF receptors 2 (VEGFR-2 [Flk1]) and 3 (VEGFR-3 [Flt4]). Interaction with VEGFR-2 results in physiological and intratumoral neoangiogenesis and vessel sprouting (Cao et al.; Tammela et al.), whereas interaction with VEGFR-3 is critical for lymphangiogenesis (Karkkainen et al.; Laakkonen et al.; Mäkinen et al.). Overexpression of VEGF-C in tumor cells has been shown to result in enhanced lymph flow and increased metastasis to regional lymph nodes (Hoshida et al.; Mandriota et al.; Padera et al.; Skobe et al.).
