Anti-MAP3K3 Rabbit Polyclonal Antibody

Supplier: Enzo Life Sciences

MEKKs (Mitogen activated protein kinase kinase kinases) are serine-threonine kinases that act as the first tier of cellular MAP kinase pathways by activation of MAP/ERK kinases, or MEKs. Many enzymes with MEKK activity have been identified, including MEKK1-4, Raf, MLK3, TAK, and DLK. MEKKs generally display little similarity outside of their catalytic kinase domains. MEKK1-4 are nearly 50% identical within their catalytic domains, and are known to regulate Erk, Jnk, and p38 MAP kinase pathways. MEKK2 and MEKK3 bind MEK5 via conserved PB1 domains, leading to downstream activation of Erk5.

Anti-JNK1 Mouse Monoclonal Antibody

Supplier: Enzo Life Sciences

JNK1 is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. JNK1 is activated by various cell stimuli, and targets specific transcription factors, and thus mediates immediate-early gene expression in response to cell stimuli. The activation of JNK1 by tumor-necrosis factor alpha (TNF-alpha) is found to be required for TNF-alpha induced apoptosis. JNK1 is also involved in UV radiation induced apoptosis, which is thought to be related to cytochrom c-mediated cell death pathway. Studies of the mouse counterpart of JNK1 gene suggested that it play a key role in T cell proliferation, apoptosis and differentiation. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

Anti-MPO Rabbit Polyclonal Antibody

Supplier: Enzo Life Sciences

Myeloperoxidase (MPO) is synthesized as a single chain precursor, which is then processed into a tetramer consisting of 2 light chains and 2 heavy chains. In response to microbial invasion, MPO is released from the cytoplasmic granules of neutrophils into the extracellular space and phagosome, where it generates hypohalous acids that have a strong microbicidal effect. Myeloperoxidase can be used as a marker of airway inflammation caused either by disease, such as asthma, or by environmental irritants.

Anti-HPN Mouse Monoclonal Antibody [clone: E4]

Supplier: Enzo Life Sciences

The Hepsin gene encodes a type II transmembrane serine protease. The encoded protein has an extracellular region that consists of two domains, a catalytic serine protease domain and a non-catalytic scavenger receptor cysteine-rich domain. This protein may be involved in diverse cellular functions including blood coagulation, maintenance of cell morphology and the growth and progression of certain cancers, particularly prostate cancer.

Anti-HSP70 Mouse Monoclonal Antibody [clone: C92F3A-5]

Supplier: Enzo Life Sciences

The Hsp70 family of heat shock proteins contains multiple homologs ranging in size from 66-78 kDa, and are the eukaryotic equivalents of the bacterial DnaK. The most studied Hsp70 members include the cytosolic stress-induced Hsp70 (Hsp72), the constitutive cytosolic Hsc70 (Hsp73), and the ER-localized BiP (Grp78). Hsp70 family members contain highly conserved N-terminal ATP-ase and C-terminal protein binding domains. Binding of peptide to Hsp70 is assisted by Hsp40, and stimulates the inherent ATPase activity of Hsp70, facilitating ATP hydrolysis and enhanced peptide binding. Hsp70 nucleotide exchange and substrate binding coordinates the folding of newly synthesized proteins, the re-folding of misfolded or denatured proteins, coordinates trafficking of proteins across cellular membranes, inhibits protein aggregation, and targets the degradation of proteins via the proteasomal pathway.

Anti-Ubiquitin Rabbit Polyclonal Antibody (DyLight® 488)

Supplier: Enzo Life Sciences

Ubiquitin is a 76-amino acid polypeptide expressed in all eukaryotic cells, highly conserved from yeast to humans. The modification of proteins by chains of ubiquitin mediates targeting of cytosolic and nuclear proteins for degradation by proteasomes. Ubiquitination represents an essential cellular process affected by a multi-enzyme cascade involving classes of enzymes known as ubiquitin-activating enzymes (E1s), ubiquitin-conjugating enzymes (E2s or Ubcs) and ubiquitin-protein ligases (E3s). E1 activates ubiquitin in an ATP-dependent manner with the formation of a thiol-ester linkage between the carboxy terminus of ubiquitin and E1. Sequential, transient thiol-ester bonds then generate between the carboxyl terminus of ubiquitin and specific cysteines of E2 and E3 enzymes. These bonds culminate in the formation of an isopeptide linkage between the activated carboxy terminus of ubiquitin and the epsilon-amino group of a lysine on a target protein, or within another ubiquitin chain (usually K48 of ubiquitin) resulting in the generation of chains of ubiquitin (polyubiquitin or multiubiquitin) from 4-20 ubiquitin moieties. Although the formation of K48-linked polyubiquitin chains on proteins constitutes a potent targeting signal for degradation in 26S proteasomes, ubiquitination is also reversible, and a number of not fully characterized deubiquitinating enzymes exist free in the cytosol. Research exploring how proteasome targets ER-retained proteins for degradation suggests that ER chaperones (such as Cne1p/calnexin, Kar2p/BiP or other components of the protein processing or transport machineries) helped by cytosolic chaperones (such as members of the Hsp70 and DnaJ families) recognize, target and force misfolded, unassembled or aberrantly modified proteins through the translocon. The retrotranslocated polypeptide chains are then released into the cytosol as soluble proteins or remain adhered to the cytosolic face of the ER membrane where deglycosylation and polyubiquitination followed by binding to the proteasome as well as degradation to peptides occurs. Ubiquitination also participates in the internalization and degradation of plasma membrane proteins such as some of the TCR subunits while still ER-membrane associated.

Anti-IgG1 Mouse Monoclonal Antibody (DyLight® 488)

Supplier: Enzo Life Sciences

Isotype matched controls are useful in flow cytometry to determine a gross estimate of non-specific binding that may be observed with a specific antibody of interest. In general, an isotype control should be of the same species and isotype as the antibody of interest, used at the same concentration, and conjugated to the same fluorophore. This reagent is an IgG1 isotype mouse monoclonal antibody (Clone # MOPC-21), conjugated to DyLight™-488. It is intended for use as an isotype control antibody for flow cytometry.

Anti-DLG4 Mouse Monoclonal Antibody

Supplier: Enzo Life Sciences

Postsynaptic density protein 95 kDa (PSD-95), also known as Synapse associated protein 90 kDa (SAP90), is a brain specific protein that is highly similar to the Drosophila dlg tumor suppressor protein. PSD-95 is a member of membrane associated proteins that are localized beneath the postsynaptic membrane of synapses in the CNS. PSD-95 interacts with NMDA receptor and Shaker-type K+ channels and contributes to their clustering and localization at the synaptic spines in hippocampal neurons and the pinceau terminal of cerebellar basket cells, respectively.
