In this experiment, students purify the restriction enzyme, Eco RI. This procedure utilised an ion exchange chromatograph step for Eco RI purification. Column fractions are assayed for the enzyme using Lambda DNA and digestion products are identified by gel electrophoresis. Fractions that contain Eco RI are identified and pooled. The total and specific activities are calculated. Recommended for advanced courses.

Ion exchange chromatography is used to separate charged molecules from complex biological samples. The charged molecules bind to a solid support carrying an opposite charge to the molecule. Proteins contain regions of charged groups on the surface which interact with the ion exchange groups immobilised on the solid support (resin column). Immobilised proteins are eluted by changing either pH or the salt gradient or a combination of both. This results in the production of white colonies amongst a background of blue colonies. This experiment provides a DNA fragment together with a linear plasmid and T4 DNA ligase. Following the ligation to synthesis the recombinant plasmid, competent E. coli cells are transformed and the number of recombinant antibiotic-resistant white and blue colonies are counted. β-galactosidase activity is assayed from blue and white bacterial cells.

The Mammalian β-Galactosidase Assay Reagent Kit is a simple and versatile assay to measure β-galactosidase activity in cultured mammalian cells. Cells grown on a plate may be lysed and tested for β-galactosidase activity by adding a single all-in-one lysis-and-assay solution. Alternatively, M-PER™ Mammalian Protein Extraction Reagent may be used to prepare lysate from washed adherent or pelleted cells, and then a portion of the lysate used to assay for β-galactosidase activity. With M-PER Reagent, there is no need to scrape cells or to perform freeze-thaw cycles.
