Deletion of the UBP43 gene in mouse leads to a massive increase of ISG15 conjugates in tissues indicating that UBP43 is a major ISG15-specific protease. UBP43 is the first ISG15-specific protease reported. Both ISG15 and UBP43 genes are known to be strongly induced by interferon, genotoxic stress, and viral infection. Hsp90 is a member of the heat shock protein 90 family that functions as a molecular chaperone and has ATPase activity. Hsp90 family proteins are highly conserved between isoforms and species. Several signal transduction pathways depend on Hsp90 function including erbB2, steroid hormone receptors (such as androgen, progesterone, glucocorticoid, and aryl-hydrocarbon), and hypoxia sensing (Hif1alpha). Recent reports show that tumor cells are more sensitive to Hsp90 inhibition and that Hsp90 from tumor cells is more sensitive to small molecule inhibitors (eg 17AAG). The mechanism of this differential sensitivity of normal versus tumor Hsp90 is not known (although mutation has been ruled out). One possible mechanism may be differences in post-translational modification of tumor Hsp90. Hsp90 is a cytoplasmic protein that forms a homodimer in vivo, and interacts with TOM34, AHSA1, HDAC6 and SMYD3.

The fusion protein consists of the N-terminus of MECT1 joined to the C-terminus of MAML2. The reciprocal fusion protein consisting of the N-terminus of MAML2 joined to the C-terminus of MECT1 has been detected in a small number of mucoepidermoid carcinomas. Multiple isoforms have been reported for the MECT1 protein.

Myosin is the major component of thick muscle filaments, and is a long asymmetric molecule containing a globular head and a long tail. Activation of smooth and cardiac muscle primarily involves pathways that increase calcium and myosin phosphorylation resulting in contraction. Myosin light chain phosphatase acts to regulate muscle contraction by dephosphorylating activated myosin light chain. The selected peptide sequence used to generate the polyclonal antibody is located near the amino terminal end of the polypeptide corresponding to the smooth/non-muscle form of myosin regulatory light chain found in cardiac myocytes in addition to smooth and non-muscle cells. This sequence differs from that of the sarcomeric/ cardiac form of myosin regulatory light chain that has a different sequence around the phosphorylation site. Human, mouse and rat have almost identical sequences.

Several Plk1-dependent phosphorylation sites have been identified on MLF1IP by mass spectrometry. Mutations of these sites stabilize MLF1IP and inhibit mitotic progression. Subsequent in vitro and in vivo MLF1IP phosphorylation and stability assays have revealed that phosphorylation of Thr78 is critical for triggering Plk1-dependent MLF1IP degradation. Expression of a non-degradable Thr78Ala mutant was sufficient to induce a mitotic block. Timely phosphorylation of MLF1IP on Thr78 by Plk1 is critical for eliminating the MLF1IP-imposed mitotic block prior to anaphase onset. MLF1IP is speculated to be a novel tumor suppressor, whose function is required for proper sister-chromatid separation. Loss of MLF1IP function may result in improper segregation of chromosomes and genomic instability, thus promoting tumorigenesis.

This protein is a ligand for the Roundabout (Robo) receptors and acts as guidance cues in axonal migration/navigation during neural development, at the ventral midline of the neural tube. Slit1 and Slit2 are essential for midline guidance in the forebrain by acting as repulsive signals preventing inappropriate midline crossing by axons projecting from the olfactory bulb. Each SLIT gene encodes a putative secreted protein, which contains conserved protein-protein interaction domains including leucine-rich repeats and epidermal growth factor-like motifs, similar to those of the Drosophila protein. In situ hybridization studies indicated that the rat SLIT-1 mRNA was specifically expressed in the neurons of fetal and adult forebrains. This data suggests that the SLIT genes form an evolutionarily conserved group in vertebrates and invertebrates, and that the mammalian SLIT proteins may participate in the formation and maintenance of the nervous and endocrine systems by protein-protein interactions. Alternative splicing isoforms have been identified for Slit1 protein.
