Recognises a sialoglycoprotein of 27 to 32 kDa, identified as CD99, or MIC2 gene product, or E2 antigen. MIC2 gene is located in the pseudo-autosomal region of the human X and Y chromosome. MIC2 gene encodes two distinct proteins, which are produced by alternative splicing of the CD99 gene transcript and are identified as bands of 30 and 32 kDa (p30/32). Although its function is not fully understood, CD99 is implicated in various cellular processes including homotypic aggregation of T cells, upregulation of T cell receptor and MHS molecules, apoptosis of immature thymocytes and leukocyte diapedesis. CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. Most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells express this antigen. Mature granulocytes express very little or no CD99. It is strongly expressed on Ewing s sarcoma cells and primitive peripheral neuroectodermal tumors.

Plasma cells differentiate from B cells upon stimulation by CD4+ lymphocytes. The B cell acts as an antigen-presenting cell (APC), consuming an offending pathogen, which is taken up by the B cell by phagocytosis and broken down within proteosomes. Plasma cells contain basophilic cytoplasm; their nucleus contains heterochromatin organized in a characteristic cartwheel arrangement. This mAb superbly recognises normal and neoplastic plasma cells in routine formalin-fixed, paraffin-embedded tissue sections. It is of potential value in identifying myeloma or plasmacytoma in bone marrow or other tissues. It also helps differentiate lympho-plasmacytoid lymphoma from lymphocytic and follicular lymphoma.

Pyruvic acid is an intermediate in sugar metabolism and in enzymatic carbohydrate degradation (alcoholic fermentation) where it is converted to acetaldehyde and CO2 by carboxylase. In muscle, pyruvic acid (derived from glycogen) is reduced to lactic acid during exertion, which is reoxidized and partially retransformed to glycogen during rest. It improves coliform recovery when present in culture medium. It is involved in a metabolic regulatory pathway activated by mitochondrial oxidants. Pyruvate is involved in respiratory regulation in plants by interacting with alternative oxidase at a conserved cysteine residue. It may help prevent hydrogen peroxide mediated cell death.
Sodium pyruvate is utilized as a component in culture broth and media. Sodium pyruvate is used in Wallen fermentation medium to enhance the conversion of oleic acid to 10-ketostearic acid by Bacillus sphaericus. Sodium pyruvate has also been used to establish stably transfected human B cell lines.
Sodium Pyruvate has shown antioxidant properties and protective effects against oxygen radicals. Pyruvate is produced as part of glycolysis and is an intermediate in many metabolic pathways. It can be converted into acetyl CoA and enter the TCA Cycle.

Anti-PNL2 is a novel monoclonal antibody, which has recently been introduced as an immunohistochemical reagent to stain melanocytes and tumors derived therefrom. The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.

The Cell-division cycle protein 20 is an essential regulator of cell division that is encoded by the CDC20 gene in humans. The major expressed form of the protein uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. This antibody recognises a protein of 205 kDa-220 kDa, identified as CD45RA, an isoform of the human leukocyte common antigen (CD45). The character of this disorder can vary, however, depending on the specific mutation since this determines age of onset and severity of symptoms.
