Dihydrorhodamine 123 is the reduced form of rhodamine 123, which is a commonly used fluorescent mitochondrial dye. It is non-fluorescent, but it readily enters most of the cells and is oxidized by oxidative species or by cellular redox systems to the fluorescent rhodamine 123 that accumulates in mitochondrial membranes. It is useful for detecting reactive oxygen species including superoxide (in the presence of peroxidase or cytochrome c) and peroxynitrite. Dihydrorhodamine 123 dihydrochloride is functionally equivalent to dihydrorhodamine 123 but with increased stability toward air oxidation and light during storage. Dihydroethidium (also called hydroethidium) is the chemically reduced form of the commonly used DNA dye ethidium bromide. The probe is useful to detect oxidative activities in viable cells, including respiratory burst in phagocytes. Dihydroethidium itself has blue fluorescence (355/420 nm) in cells, while the oxidized form ethidium has red fluorescence (518/605 nm) upon DNA intercalation. H2DCFDA (2′,7′-Dichlorodihydrofluorescein diacetate) is a useful fluorogenic reagent to detect reactive oxygen intermediates in cells. On oxidation, H2DCFDA becomes the highly green fluorescent 2′,7′-dichlorofluorescein, 504/529 nm (end product).

EDC reacts with a carboxyl group first and forms an amine-reactive O-acylisourea intermediate that quickly reacts with an amino group to form an amide bond with release of an isourea by-product. The intermediate is unstable in aqueous solutions and so two-step conjugation procedures rely on N-hydroxysuccinimide (NHS) for stabilization. ORAI3 Monoclonal Antibody: Antigen stimulation of immune cells triggers Ca++ entry t hrough Ca++ release-activated Ca++ (CRAC) channels. ORAI3 is one of two mammalian homologs to ORAI1, a recently identified four-transmembrane spanning protein that is an essential component of CRAC. All three homologs have been shown to function as Ca++ plasma membrane channels gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca++ sensor. However, ORAI3 channels failed to produce detectable Ca++ selective currents in cells co-transfected with ORAI3 and STIM1, indicating that ORAI3 channels undergo a lesser degree of depotentiation than ORAI1 or ORAI2. Na+ currents through ORAI1, 2 and 3 channels were equally inhibited by extracellular Ca++, indicating that each have similar affinities for Ca++ within the selectivity filter. This antibody is predicted to have no cross-reactivity to ORAI1 or ORAI2. Larger molecular weight bands are sometimes seen in SDS-PAGE; these may represent post-translationally modified ORAI 3.
