5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside, commonly known as X−Gal, is a histochemical substrate for β−galactosidase.
5-Bromo-4-Chloro-3-Indolyl-β-D-Galactopyranoside is used as indigogenic substrate for β-galactosidase, for detection of β-galactosidase-positive clones, and the identification of lac and bacterial colonies or phage plaques. It is the substrate of choice for blue-white selection of recombinant bacterial colonies with the lac+ genotype. X−Gal is cleaved by β−galactosidase to yield an insoluble blue precipitate. X−Gal is particularly useful in molecular biology applications to detect the activity of β−galactosidase which is frequently used as a reporter gene. In cloning, X−Gal is used to detect insertion of foreign DNA into the lacZ region of plasmid DNA using α-complementation which is based on vectors such as the pUC and the M13mp series that carry a fragment of the β-galactosidase gene encoding an α-fragment of β-galactosidase.

T-Cell Intracellular Antigen-1 is a cytoplasmic granule-associated protein expressed in lymphocytes processing cytolytic potential. It is a member of an RNA-binding protein family and possesses nucleolytic activity against cytotoxic lymphocyte (CTL) target cells. It has been suggested that this protein may be involved in the induction of apoptosis as it preferentially recognises poly(A) homopolymers and induces DNA fragmentation in CTL targets. The major granule-associated species is a 15 kDa protein thought to be derived from the carboxyl terminus of the 40 kDa product by proteolytic processing. TIA-1 antibody labels cytotoxic T cells and natural killer cells (NK cells). It is also expressed in T-cell lymphoma, large granular lymphocyte (LGL) leukemia and hairy cell leukemia. TIA1 expression in T-cell malignancies may help in differentiating LGL leukemia (high expression) from T-cell lymphocytosis and other T-cell diseases (low expression). TIA1 may also be used to label tumor-infiltrating lymphocytes in the study of immune response to malignancies.

Heparanase is an endo-beta-D-glucuronidase, which degrades heparan sulfate side chains of heparan sulfate proteoglycans (HSPGs) in the extracellular matrix. Heparanase plays an important role in ECM degradation, facilitating the migration and extravasation of tumor cells and inflammatory leukocytes. Upon degradation, heparanase releases growth factors and cytokines that stimulate cell proliferation and chemotaxis. Heparanase is a heterodimer comprised of a 50k D subunit harboring the active site and a 8kD subunit. It is produced as a latent 65 kD precursor and proteolytically processed to its active form. Heparanase is highly expressed in myeloid leukocytes (i.e. neutrophils) in platelets and in human placenta. Human heparanase was found to be upregulated in various types of primary tumors, correlating in some cases with increased tumor invasiveness and vascularity and with poor prospective survival. Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. Reacts with the 65 kD precursor as well as the 50 kD and 8 kD subunits of human or mouse Heparanase.
