The activating transcription factor ATF-2 (also called CRE-BP1) binds to both AP-1 and CRE DNA response elements and is a member of the ATF/CREB family of leucine zipper proteins. ATF-2 has been implicated in the transcriptional regulation of a number of genes including cytokines, cell cycle control and apoptosis. Various forms of cellular stress, including inflammatory cytokines and UV irradiation, stimulate the transcriptional activity of ATF-2. 14-3-3 proteins are a family of highly conserved proteins that appear to have multiple roles in cell signaling (Bridges and Moorhead, 2005). The proteins are abundantly expressed in the brain and have been detected in the cerebrospinal fluid of patients with different neurological disorders (Berg et al., 2003). 14-3-3 proteins bind protein ligands that are typically phosphorylated on
serine or threonine residues and regulate the functions of these binding partners by a number of different mechanisms (Silhan et al., 2004; Dougherty and Morrison, 2004). The14-3-3 proteins affect a diverse array of cellular processes including the cell cycle and transcription, signal transduction and intracellular trafficking. The transcription factor Forkhead-box A1 (FOXA1), also known as hepatocyte nuclear factor 3-alpha, is a member of the FOX class of transcription factors. HNF-1 (alpha and beta), HNF-3 (alpha, beta and gamma), HNF-4 (alpha and gamma), and HNF-6 compose, in part, a homeoprotein family designated the hepatocyte nuclear factor family. FOXA1 is expressed in normal breast ductal epithelium and other epithelium in different organs, such as lung, pancreas, bladder, prostate, and colon. Recently, FOXA1 has been shown to be a major determinant of estrogen-ER activity and endocrine response in breast cancer cells. FOXA1 expression correlates with estrogen receptor (ER)-positivity, especially in luminal subtype A breast cancers, which is associated with favorable prognosis. PECAM-1, or platelet endothelial cell adhesion molecule-1, also called CD31, is made up of six extracellular immunoglobulin-like domains, a single transmembrane domain, and a cytoplasmic domain containing serine and tyrosine residues that when phosphorylated, activate the molecules signaling function. It is expressed on platelets, monocytes, neutrophils, bone marrow macrophage and dendritic cells precursors, and at the intracellular junction of endothelial cells. PECAM-1, on the endothelial lining of blood vessels, helps maintain the vascular barrier. In response to inflammatory mediators, the endothelial pull away from each other allowing fluid, inflammatory cells and proteins into the extravascular space. The inflammatory response fades as adhesion of neighboring cells increase. PECAM-1 also plays an important role in angiogenesis, promoting endothelial cell motility, junctional development and capillary morphogenesis.

This gene is a member of the protocadherin alpha gene cluster, one of three related gene clusters tandemly linked on chromosome five that demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The alpha gene cluster is composed of 15 cadherin superfamily genes related to the mouse CNR genes and consists of 13 highly similar and 2 more distantly related coding sequences. The enzyme 5-lipoxygenase (5-LO) plays a key role in regulating the production of leukotrienes (LTs) (Funk, 2001). Overproduction of LTs contributes to several diseases, most notably chronic inflammatory diseases, including asthma (Drazen et al., 1994), fibrosis (Wilborn et al., 1996) and atherosclerosis (Dwyer et al., 2004). Recent work has demonstrated that the activity of 5-LO is regulated by PKA phosphorylation of serine-523 in 5-LO (Luo et al., 2004). Under normal conditions, this phosphorylation may be important in limiting inflammation. Abnormal signaling through cAMP and PKA, then, could contribute to a variety of diseases, including those characterized by chronic inflammation. The phospho-specific antibody to Ser523 on 5-LO is thus likely to provide a valuable tool for studies of the role of 5-LO regulation in diseases such as asthma, fibrosis and atherosclerosis.

Reportedly, CD3 complex is involved in signal transduction to the Tcell interior following antigen recognition. Beta-catenin associates with the cytoplasmic portion of E-cadherin, which is necessary for the function of E-cadherin as an adhesion molecule. CD31, also called PECAM-1, is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. It is expressed by stem cells of the hematopoietic system and CD31 antibody can be used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. CD31 antibody has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. This antibody reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high expression level may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.
