Receptor activator of nuclear factor kappa-B ligand (RANKL), also known as tumor necrosis factor ligand superfamily member 11 (TNFSF11), is a protein that in humans is encoded by the TNFSF11 gene. This gene encodes a member of the tumor necrosis factor (TNF) cytokine family which is a ligand for osteoprotegerin and functions as a key factor for osteoclast differentiation and activation. It is mapped to chromosome13q14.11. Targeted disruption of the related gene in mice led to severe osteopetrosis and a lack of osteoclasts. The deficient mice exhibited defects in early differentiation of T and B lymphocytes, and failed to form lobulo-alveolar mammary structures during pregnancy. This gene may play an important role in enhanced bone-resorption in humoral hypercalcemia of malignancy.

Mutations in this gene result in familial dysbetalipoproteinemia, or type III hyperlipoproteinemia (HLP III), in which increased plasma cholesterol and triglycerides are the consequence of impaired clearance of chylomicron and VLDL remnants. Alternative splicing results in multiple transcript variants.

Programmed death-ligand 1 (PD-L1), also known as CD274 or B7-H1, is a protein that in humans is encoded by the CD274 gene. It is mapped to 9p24.1. PD-L1 is a 40kDa type 1 transmembrane protein that has been speculated to play a major role in suppressing the immune system during particular events such as pregnancy, tissue allografts, autoimmune disease and other disease states such as hepatitis. It has been concluded that upregulation of PD-L1 on tumor MDCs downregulates T-cell immunity and that PD-L1 blockade may represent an approach for cancer immunotherapy. Additionally, PD-L1 can provide positive costimulatory signals for innate and adaptive immunity and for protection against intracellular bacterial infection. This gene encodes a member of the non-histone chromosomal high mobility group protein family.
