The procedure for labeling of DNA probes with a polynucleotide 'tail' containing hapten-labeled nucleotides was developed by Enzo. In such terminal labeling reactions, terminal transferase catalyzes the addition of nucleotides to any 3'-OH terminus in a template independent manner. This rapid and convenient nonradioactive labeling procedure is free of any sequence bias that is normally observed in random priming or nick translation reactions. The Cytomegalovirus (CMV) BioProbe® labeled probe is prepared by nick translation of cloned fragments of the CMV Towne strain. The fragments include a total of 30-31 kb of DNA, approximately 20% of the CMV genome. The probe is specific for cytomegalovirus DNA. It does not hybridize to DNAs of other herpesviruses (i.e., herpes simplex virus or Epstein-Barr virus) but it has been found to hybridize to the DNA of CMV clinical isolates. Fragment size range: 100-1000 base pairs (as estimated by agarose gel electrophoresis).

The Adenovirus BioProbe® labeled probe is prepared by nick translation of cloned fragments of the adenovirus 5 genome. The fragments constitute a total of 20 kb of DNA, approximately 56% of the genome. The probe is specific for adenovirus and, in addition to hybridization to adenovirus 5 sequences, the probe will cross hybridize with adenovirus types 4, 7, 11, 20, 40 and 41 due to their considerable sequence homology.
