Protein Linking and Labeling Systems

Protein linking and labelling systems provide a convenient means for attaching fluorescent labels or biotins to antibodies and larger proteins. These kits are ideal for use with multiple applications, including flow cytometry, fluorescent microscopy, immunohistochemistry, primary detection, ELISAs, immunocytochemistry, and FISH. These systems are available with a variety of dye colours, biotin, fluorescent agents, and all the components needed to perform multiple conjugations and purifications. Protein labelling follows a simple protocol of reaction, separation, and then use.

Phosphotyrosine detection set

Supplier: ENZO LIFE SCIENCES

This kit offers a set of seven different MAbs specific for tyrosine phosphorylation sites. Phosphorylation patterns in a given cell extract may differ when probed with different antibodies due to sequence specificity. Recognition therefore depends on two criteria: Phosphorylation and surrounding aa motif. If one of the two criteria is not fulfilled, the antibody will not detect the phosphorylation site. Specificity has been determined by epitope mapping using degenerated phosphopeptide libraries.

Phalloidin conjugates

Phalloidin conjugates

Supplier: Biotium

Phalloidin is a toxin isolated from the deadly Amanita phalloides mushroom. It is a bicyclic peptide that binds specifically to F-actin. Fluorescent conjugates of phalloidin are convenient tools to investigate the distribution of F-actin in fixed and permeabilised cells or tissue sections. One unit (U) of phallodin is sufficient to stain one sample (200 µl staining volume). A highly selective actin filament stain for fixed and permeabilised cells. Labeled phalloidins have similar affinity for both large and small filaments.

Lipophilic carbocyanine dyes and CellBrite cytoplasmic membrane labelling kits

Supplier: Biotium

Carbocyanine dyes with long hydrocarbon chains have low fluorescence in aqueous phase, but become highly fluorescent upon binding to lipid bilayers, making this class of dyes ideal for staining cell membranes. Carbocyanine dye membrane staining is very stable, with minimal dye transfer between cells. Dilinoleyl carbocynaine dyes have much faster lateral diffusion rates than regular carbocyanine dyes, which facilitates labelling of neurons in tissues.

Orange 552 dUTP

Supplier: ENZO LIFE SCIENCES

Orange 552 dUTP (5-TAMRA-dUTP) can replace TTP in reactions in which it serves as a substrate for E. coli DNA polymerase (holoenzyme and Klenow fragment), T4 and Taq DNA polymerases, reverse transcriptase (from AMV and M-MuLV) and terminal transferase. Fluorescently labeled probes can be prepared with this fluorescent nucleotide by a variety of methods including nick translation, cDNA labeling and 3’-end labeling. Probes generated by these methods are suitable for use for the identification of specific sequences by in situ hybridization procedures on fixed cells and tissues by direct fluorescence detection. Orange 552 dUTP can also be used for multicolor fluorescence labeling.
