Vectors, Plasmids and Libraries

In molecular cloning, vectors are DNA molecules used to artificially deliver foreign genetic material into another cell. The four major types of vectors are plasmids, viral vectors, cosmids, and artificial chromosomes, with the most commonly used of these being plasmids. Vectors may be used for cloning, and certain vectors are specifically tailored to that exact purpose, while others may be designed specifically for transcription and protein expression. Simpler transcription vectors are used to amplify their inserted genetic material.

Vector, pBR322, MP Biomedicals

Supplier: MP Biomedicals

pBR322 is a 4363 bp vector purified from Escherichia coli. It is one of the most commonly used cloning vectors, that confers resistance to ampicillin and tetracycline. It contains single restriction sites for Pst I and Pvu I within the ampicillin-resistance gene (ampr) and single restriction sites for BamH I, Sal I, Sph I, EcoR V, Nru I and Xma III within the tetracycline-resistance gene (tetr).

Gaussia Luc vector for luciferase assays, expression vectors, Pierce™

Gaussia Luc vector for luciferase assays, expression vectors, Pierce™

Supplier: Thermo Fisher Scientific

Gaussia Luc vectors contain a gene cloned from the copepod, Gaussia princeps. The gene encodes a naturally-secreted bioluminescent Gaussia luciferase (approx. 20 kDa), which enables measurement of the reporter activity in media (for real-time assays) and in cell lysates.

Pierce™ Gaussia-Dura Luc Vector for Luciferase Assays, Expression Vectors

Pierce™ Gaussia-Dura Luc Vector for Luciferase Assays, Expression Vectors

Supplier: Thermo Fisher Scientific

Gaussia Dura Luc vectors contain a mutant form of the Gaussia luciferase gene that confers better bioluminescent signal stability than native luciferase. Gaussia-Dura luciferase (approx. 20 kDa) is a secreted protein, which enables measurement of the reporter activity in media (for real-time assays) and in cell lysates.

Expression vectors, pGEX

Supplier: Cytiva

The pGEX vectors have an expanded multiple cloning site (MCS) that contains six restriction sites. The expanded MCS facilitates the unidirectional cloning of cDNA inserts obtained from libraries constructed using many available lambda vectors. pGEX-6P-1, pGEX-6P-2, and pGEX-6P-3 each encode the recognition sequence for site-specific cleavage by PreScission Protease between the GST domain and the multiple cloning site. pGEX-4T-1, pGEX-4T-2, and pGEX-4T-3 are derived from pGEX-2T and contain a Thrombin recognition site. pGEX-5X-1, pGEX-5X-2, and pGEX5X-3 are derivatives of pGEX-3X and possess a Factor Xa recognition site.
