Fast green FCF, Certistain® for microscopy, Sigma-Aldrich®

Fast green FCF, Certistain® for microscopy, Sigma-Aldrich®

Supplier: Merck

The Certistain® dry dye Fast Green FCF (C.I. 42053) is used for the preparation of a staining solution that can be used for modified connective tissue staining acc. to van Gieson in histological sections. Further it is applied in histology as counterstaining in e.g. trichrome staining acc. to Masson and additionally for histochemical staining, as well as in Cytology as counterstain instead of light green SF in Papanicolaou's staining.
As CERTISTAIN® dye it is chemically quality-certified according to highest standards and strict specifications. As an IVD product and CE registered, it can be used hassle-free in diagnostics and laboratory accreditations.

Ethidium homodimer I used in nuclear staining

Supplier: Biotium

Ethidium Homodimer, first developed by Dr. Le Pecq and his colleagues, is a high-affinity fluorescent nucleic acid stain. It binds to both DNA and RNA in a sequence-independent manner and with a >30-fold fluorescence enhancement. The DNA binding of each Ethidium Homodimer covers four base pairs and is believed to occur by intercalation. Because the dye is highly positively charged, it cannot cross cell membranes to stain living cells. It is useful for detecting nucleic acids in solution, or for selectively staining dead cells with damaged plasma membranes.

Trypan blue 0.4% in PBS, dark blue solution

Trypan blue 0.4% in PBS, dark blue solution

Supplier: MP Biomedicals

Trypan Blue is a blue acid dye with a strong affinity for cellulose containing substrates such as cotton; less affinity for proteinaceous materials. Trypan blue solution may be used in trypan blue based cytotoxicity and proliferation assays.
Trypan Blue is used as a vital dye which is especially important because it is taken up by the reticuloendothelial system.

Zinc Formalin Fixative

Supplier: SIGMA ALDRICH MICROSCOPY

Zinc formalin fixative is a non-precipitating fixative. This fixative is compatible with histochemical, immunochemical and special stains. It may be used with automated tissue processors and also in manual methods. The zinc chloride component of the formulation increases the rigidity of cellular components to withstand subsequent processing, sectioning and staining procedures. Tissue specimens initially fixed using 10% neutral buffered formalin solution may be post-fixed using the zinc formalin fixative.

Histo/Zyme

Supplier: SIGMA ALDRICH MICROSCOPY

Histo/Zyme is a stabilized proteolytic enzyme in buffered solution, used to unmask immunoreactive sites altered by fixation and/or the embedding process. Treatment with a proteolytic enzyme in immunohistochemistry (IHC) is required to digest excess linkages and expose the antigen. The use of Histo/Zyme results in superior staining over traditionally used enzyme pretreatments, such as pepsin and trypsin. Moreover, Histo/Zyme requires only a brief incubation at room temperature. Performing antigen recovery with Histo/Zyme will allow you to dilute the primary antibody 5 to 10 fold further and achieve optimal staining.
