Giemsa’s stain is frequently used for diagnostic purposes in hematology to differentiate the nuclear and cytoplasmic morphology of platelets, RBCs, WBCs, and parasites. It is frequently used in combination with other dye solutions as May-Grunwald’s solution for Pappenheim (MGG) and Wright-Giemsa. The Giemsa stain was adapted to histology due to its unique staining of chromatin, nuclear membranes, and cytoplasmic elements. The reduced form of MPMS converts tetrazolium salt to purple precipitate formazan. It is a very stable and non-enzymic electron carrier that mediates electron transfer by dehydrogenases in vitro to various electron acceptors such as tetrazolium dyes or the electrode of an enzymic electric cell.

3,3′-Diaminobenzidine has been used as a chromogen for the visualisation in immunohistochemistry of chondrosarcoma sections, mammary carcinomas and brain tissue sections. Peroxidase substrate; reagent for spectrophotometric determination of selenium. DAB (3,3′-Diaminobenzidine) is utilized in many applications for the visualisation of peroxidase activity. In the peroxidase reaction, DAB serves as a hydrogen donor in the presence of peroxide. The oxidised DAB forms an insoluble brown end-product for use in the immunohistological and immunoblotting staining procedures.
