MTT, XTT, and resazurin are reduced by mitochondrial metabolic activity to yield coloured or fluorescent products, and thus are useful for and assaying cell viability and quantitating cell number. MTT and XTT are reduced to coloured formazin salts that can be measured by absorbance. MTT generates an insoluble formazin salt, requiring cell lysis before the absorbance can be measured, while XTT does not require cell lysis for measurement. Resazurin is a non-fluorescent blue dye that is reduced to the pink fluorescent compound resorufin, which can be measured by fluorescence or absorbance.

This antibody recognizes CD16 (FcγRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50-80 kDa. Two similar genes represent CD16, CD16A (FcγRIIIA), which exists as a hetero-oligomeric polypeptide-anchored form in macrophages and NK cells and CD16B (FcγRIIIB), which exist as a monomeric GPI-anchored form in neutrophils. Furthermore, there are two known polymorphisms of CD16B, NA-1 and NA-2. Individuals homozygous for NA-2 show a lower phagocytic capacity compared with NA-1. CD16 binds IgG in the form of immune complexes and shows preferential binding of IgG1 and IgG3 isotypes and minimal binding of IgG2 and IgG4. Upon IgG binding, both CD16 isoforms initiate signal transduction cascades that lead to a variety of responses including antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, degranulation and proliferation.

This antibody recognizes a ~90-95 kDa protein which is identified as cell surface transferrin receptor (CD71), a disulfide-bonded homodimeric glycoprotein of 180-190 kDa. This MAb is highly specific to CD71 and shows no cross-reaction with other related proteins. Its epitope is localized in the extracellular domain of CD71. Ligand for transferrin receptor is the serum iron transport protein, transferrin. This receptor is broadly distributed in carcinomas, sarcomas, leukemias, and lymphomas. CD71/Transferrin receptor has been reported to be associated with cell proliferation in both normal and neoplastic tissues and useful in predicting clinical behavior or response to therapy in a number of malignancies including breast cancer.

CD1 proteins have been demonstrated to restrict T cell response to non-peptide lipid and glycolipid antigens and play a role in non-classical antigen presentation. CD1a is a non-polymorphic MHC Class 1 related cell surface glycoprotein, expressed in association with Beta-2 microglobulin. Anti-CD1a labels Langerhans cell histiocytosis (Histiocytosis X), extranodal histiocytic sarcoma, a subset of T-lymphoblastic lymphoma/leukemia, and interdigitating dendritic cell sarcoma of the lymph node. When combined with antibodies against TTF-1 and CD5, anti-CD1a is useful in distinguishing between pulmonary and thymic neoplasms since CD1a is consistently expressed in thymic lymphocytes in both typical and atypical thymomas, but only focally in 1/6 of thymic carcinomas and not in lymphocytes in pulmonary neoplasms. This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. TRIM29 is also able to distinguish lung squamous cell carcinoma from lung adenocarcinoma with ~90% positive accuracy, when used in a panel with TTF-1, p63, CK5/6, and Napsin-A antibodies.

This antibody recognizes a 45 kDa protein, which is identified as isocitrate dehydrogenase (IDH1). It belongs to the isocitrate and isopropylmalate dehydrogenases family. IDH1 catalyzes the third step of the citric acid cycle, which involves the oxidative decarboxylation of isocitrate, formingalpha-ketoglutarate and CO2 in a two-step reaction. The first step involves the oxidation of isocitrate to the intermediate oxalosuccinate, while the second step involves the production ofalpha-ketoglutarate. During this process, either NADH or NADPH is produced along with CO2. Recently, an inactivating mutation of IDH1 has been implicated in glioblastoma. IDH1 appears to function as a tumor suppressor that, when mutationally inactivated, contributes to tumorigenesis in part through induction of the HIF-1 pathway.

Recognizes a protein of 150 kDa, which is identified as the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120-150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70-80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This MAb recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction.

This antibody recognizes a 63 kDa protein, which is identified as TOX3. It contains a high mobility group (HMG)-box, which regulates Ca2 -dependent transcription in neurons through interaction with the cAMP-response-element-binding protein (CREB). TOX3 appears to be associated with breast cancer susceptibility and is expressed downstream of a cytoprotective cascade together with CITED1, a transcriptional regulator that does not bind directly to DNA. TOX3 is predominantly expressed in the brain and forms homodimers. TOX3 overexpression protects neuronal cells from cell death caused by endoplasmic reticulum stress or BAX overexpression through the induction of anti-apoptotic transcripts and repression of pro-apoptotic transcripts.
