Serum- and BPE-free medium for differentiation of human primary bronchial epithelial cells or human airway epithelial cells to mature apical-out airway organoids.

Generate apical-out airway organoids in 15 days from human bronchial epithelial cells (HBECs) or human airway epithelial cells (HAECs) with a simple and scalable, Matrigel®-free protocol. These organoids provide access to the apical side of the airway epithelium and can be used to perform infectious disease modeling or high-throughput drug screening in an in vitro, three-dimensional (3D) model system. PneumaCult™ Apical-Out Airway Organoid Medium allows growth of polarised airway organoids with outward-facing, differentiated ciliated cells in AggreWell™400 24-well plates - without the use of cell culture inserts. To maximise experimental reproducibility, this medium is formulated without serum or BPE. For a complete apical-out airway organoid culture workflow, expand HBECs or HAECs in PneumaCult™-Ex Plus Medium prior to initiating organoid culture with PneumaCult™ Apical-Out Airway Organoid Medium.
