SARS-CoV-2 Recombinant Spike Protein, aa319-541 (Yeast-expressed), His Tag
Supplier: STEMCELL Technologies
SARS-CoV-2 Recombinant Spike Protein, aa319-541 is expressed in Pichia pastoris and is one of four structural proteins encoded by the SARS-CoV-2 genome. The Spike Protein plays a key role in attachment to host cells allowing invasion through clathrin-mediated endocytosis. The spike protein can be cleaved by host cell proteases after aa685 to yield the N-terminal S1 subunit, and C-terminal S2 region. The S1 subunit is responsible for interacting with the host cell receptor (angiotensin-converting enzyme II) through a receptor-binding domain that is highly conserved with SARS-CoV. The S1 subunit has two conformations: a ‘down’ conformation in which the receptor is inaccessible, and an ‘up’ conformation in which the receptor is accessible. These conformational changes are key for monoclonal antibody drugs and vaccine development. At the amino terminus of the polypeptide chain, SARS-CoV-2 Recombinant spike protein contains a polyhistidine tag and a SUMOstar site.

Human Recombinant ACE2 Protein, aa18-740 (HEK293-expressed)
Supplier: STEMCELL Technologies
Human Recombinant Angiotensin-Converting Enzyme 2 (ACE2) Protein, aa18-740, is expressed in HEK293 cells. ACE2 activity results in vasodilation by degrading vasoconstrictor angiotensin II to produce vasodilator angiotensin. Decreased expression of this enzyme is associated with cardiovascular disease and possibly also glomerular injury in diabetic nephropathy. ACE2 contains a peptidase domain at the N-terminus and a single transmembrane helix. ACE2 is also the primary human receptor for the surface spike glycoprotein of SARS-CoV-2 and SARS-CoV. This receptor is expressed in human lung and small intestine epithelia as well as the heart, kidney, and testes. The peptidase domain of ACE2 binds to the protein-binding domain of the S1 protein subunit of the spike protein; this binding results in a cleavage site becoming exposed. Cleavage, or ‘protein priming’, is done by TMPRSS2, a cellular serine protease, and by cathepsins. This cleavage activates the S2 subunit of the spike protein, allowing the fusion of the viral membrane to the host cell membrane. At the carboxy terminus, Human Recombinant ACE2 Protein contains a TEV site and a human IgG1 Fc tag.

Human Recombinant Apolipoprotein A-I, His tag
Supplier: STEMCELL Technologies
A member of the apolipoprotein family, Apolipoprotein A-I (apo A-I) is a major glycoprotein component of high density lipoprotein (HDL) particles (Frank and Marcel) which facilitate the reverse transport of cholesterol from peripheral tissues to the liver. Apo A-I binds to the scavenger receptor SR-A1, allowing HDL particles to interact with cells in peripheral tissues (Neyen et al.), and is involved in the esterification of cholesterol, acting as a cofactor for lecithin cholesterol acyltransferase (LCAT; Frohlich). Apo A-I has been shown to bind to the scavenger receptor SR-B1 (Williams et al.) and the beta chain of ATP synthase (Martinez et al.) on hepatocytes, highlighting its importance in HDL endocytosis. Mutations in the APOA1 encoding gene may result in dysregulated HDL levels, leading to an increased risk of amyloidosis and ischemic heart disease (Haase et al.; Obici et al.). This protein product contains a His-residue tag at the amino end of the polypeptide chain. This protein product contains a His-residue tag at the amino end of the polypeptide chain. For consistency and reproducibility across your applications, sclerostin from STEMCELL comes lyophilised with ≥92% purity.

Human Recombinant ASAM, His tag
Supplier: STEMCELL Technologies
Adipocyte-specific adhesion molecule (ASAM) may play a role in cell-cell adhesion as it is expressed in epithelial cells of various tissues and is associated with tight junctions (Raschberger et al., Sze et al.). This type 1 transmembrane protein belongs to the cortical thymocyte marker in the Xenopus (CTX) family within the immunoglobulin superfamily. Sequence alignment analysis has shown that ASAM shares conserved characteristics with other CTX-like proteins, including extracellular V-type and C2-type domains, a transmembrane region, and a cytoplasmic tail (Raschperger et al.). ASAM is also expressed in adipocytes, and was found to be upregulated in white adipose tissue (WAT) in a rodent model of obesity and type II diabetes (Eguchi et al.). This protein contains a His-residue tag at the carboxyl end of the polypeptide chain. For consistency and reproducibility across your applications, adipocyte-specific adhesion molecule from STEMCELL comes lyophilised with ≥94% purity, and is verified by LAL analysis to ensure endotoxin levels are ≤1,0 EU/μg protein.

Mouse Recombinant IFN beta
Supplier: STEMCELL Technologies
A cytokine belonging to the type 1 interferon family, Interferon beta (IFN beta) binds IFN alpha/beta receptors (IFNAR) that activate tyrosine kinases and initiate the interferon-induced Jak-STAT signaling pathway, which modulates many key immune processes (Smieja et al.). In an experimental model involving cardiac fibroblasts isolated from rats, IFN beta was found to induce both pro- and anti-inflammatory cytokines production by activating different STAT proteins (Bolivar et al.). The anti-inflammatory effects of IFN beta have been studied in the context of autoimmune disorders, and there are currently multiple approved IFN beta drugs for treatment of relapsing forms of multiple sclerosis (Filipi and Jack). IFN beta is produced by immune cells, including macrophages, and non-immune cells, such as fibroblasts and epithelial cells (Ivashkiv and Donalin). The crystal structure of IFN beta shares characteristics with other type I interferons. It comprises five alpha-helices with four of them forming a helix bundle, and one long and three shorter loops connecting the helices (Karpusas et al.). For consistency and reproducibility across your applications, interferon alpha 1 from STEMCELL comes lyophilised with ≥87% purity, specific activity EC50 ≤18 pg/mL, and LAL analysis verification ensuring endotoxin levels are ≤1,0 EU/μg protein.

Human Recombinant IFN alpha 1, His Tag
Supplier: STEMCELL Technologies
Interferon alpha 1 (IFNA1) belongs to the type 1 interferon family of cytokines, which bind interferon alpha receptors (IFNAR; composed of IFNAR1 and IFNAR2 subunits) that are involved in interferon-induced JAK-STAT signaling (Shemesh et al.). Interferons have inhibitory effects on viral replication and pathogenesis, and studies have found that IFN alpha can prevent the spread of herpes simplex virus (HSV) (Mikloska and Cunningham), and human immunodeficiency virus (HIV) (George and Mattapallil). Pegylated forms of IFN alpha are currently approved for the treatment of chronic hepatitis B (Woo et al.). IFN alpha also displays anti-tumor effects by regulating cell growth and proliferation, and it is used in the treatment of different cancers (Tagliaferri et al.). Interferons display alpha-helical structures with four helices forming an antiparallel alpha-helix bundle (Walter). This protein contains a His-residue tag at the carboxyl end of the polypeptide chain. For consistency and reproducibility across your applications, interferon alpha 1 from STEMCELL comes lyophilised with ≥90% purity, specific activity EC50 ≤40 to 200 pg/ml, and LAL analysis verification ensuring endotoxin levels are ≤1,0 EU/μg protein.
