HiPrep™ Heparin FF 16/10 is prepacked with Heparin Sepharose 6 Fast Flow for preparative purification of proteins with affinity for heparin. Heparin is a mucopolysaccharide consisting of a repeating dimer of α-L-dipyranuronic acid 2-sulfate, and 2-deoxy-2-sulfamino-α-D-glucopyranose 6-sulfate, with a molecular weight range of 5000 to 30 000.

  • 20 ml column optimised for convenient scale-up purification Immobilized heparin is used for purification of antithrombin III, coagulation factors, and other plasma proteins lipoproteins, protein synthesis factors, enzymes that act on nucleic acids, and steroid receptors Simple operation with single pump configurations, as well as ÄKTA design systems Excellent flow characteristics

Heparin functions as a high-capacity cation exchanger, due to its anionic sulfate groups. Gradient elution with salt is most commonly used. Heparin is coupled to Sepharose 6 Fast Flow with a chemically optimized linkage.

Heparin Sepharose 6 Fast Flow is composed of cross-linked 6% agarose beads, modified with sodium heparin. Immobilization is performed using chemically stable and oriented coupling, which delivers high binding capacity. The chemical stability of the heparin resin is solely dictated by the ligand.
