Minimum Essential Medium (MEM) with Earle's Balanced Salts is a modification of Eagle's earlier medium, Basal Medium Eagle (BME), containing higher concentrations of the essential nutrients. These media have demonstrated the ability to support a variety of normal and transformed cells in culture and contain Earle's Balanced Salts, which make them suitable for use in atmospheres charged with CO₂ gas.

In 1976, Guilbert and Iscove demonstrated that precursor cells of erythrocytes and macrophages could be cultured in a reduced-serum medium supplemented with albumin, transferrin, lecithin, and selenium. Iscove's medium is a modification of Dulbecco's Modified Eagle's Medium (DME) containing selenium, additional amino acids and vitamins, sodium pyruvate, HEPES buffer, and potassium nitrate instead of ferric nitrate. Further studies have demonstrated that Iscove's medium will support murine B lymphocytes, haemopoietic tissue from bone marrow, B-cells stimulated with lipopolysaccharide, T-lymphocytes, and a variety of hybrid cells.

McCoy’s media were originally formulated for growth and support of lymphocytes. This final modification produced a medium identical to RPMI 1629. McCoy's 5A media supports the indefinite proliferation of Walker 256 carcinoma cells. In addition, it is excellent for the propagation of leukocytes, biopsy tissues, a broad range of human and rat normal or transformed cell types, the most current primary and continuous cell lines.

These irrigating buffers and salt solutions are sterile, physiological balanced solutions intended for use in the maintenance of mammalian cells, where a chemically defined, balanced salt solution provides an environment that will maintain the structural and physiological integrity of cells in vitro. Applications include the intermediate steps for cultivation of cells, like washing, centrifugation, suspending and counting, as well as for many analytical methods or biochemical treatments. Principally these solutions should be used to keep the cells in suspension only for minutes or a few hours maximum. These buffers and solutions are not cell culture media.
