The E.Z.N.A.® HP Total RNA Isolation Kit provides a rapid and easy method for RNA isolation from a small amount of cultured eukaryotic cells or tissues. There is no need for phenol/chloroform extractions, time-consuming steps such as CsCl gradient ultracentrifugation, or precipitation with isopropanol or LiCl. RNA purified using the E.Z.N.A.® HP Total RNA method is ready for applications such as RT-PCR, RT-qPCR, Northern blotting, poly A+ RNA (mRNA) purification, nuclease protection, and in vitro translation.

Modified from the E.Z.N.A.® Plant RNA kit, the E.Z.N.A.® Fungal RNA kit uses an improved buffer system that is more efficient for fungal samples. This kit provides convenient and rapid methods for the isolation of total RNA from a variety of fungal species. Rather, the method involves a simple and rapid precipitation step for removal of much of the polysaccharides and phenolic compounds commonly found in fungal tissues. In combination with HiBind® RNA spin columns, this method permits purification of high quality RNA from as much as 200 mg tissue. The system is also efficient enough to purify total RNA from as little as 10 mg of tissue or 100 cells. Purified RNA can be used for many downstream applications such as RT-PCR, northern blotting, Poly(A)+ mRNA purification, and differential display.

The sample is first lysed and homogenised in a special denaturing buffer, spun to pellet RNA and undigested particles, and then the supernatant is applied to a HiBind® DNA spin column to bind DNA. The RNA pellet is dissolved and purified by a HiBind RNA spin column. Since there is no need to divide the sample into two parts for separate purification procedures, maximum yield of DNA and RNA can be purified from the entire sample.
