Medium 199 was developed specifically for nutritional research of chicken fibroblasts. Today, 199 media are widely used for the maintenance of non transformed cells, vaccine and virus production and primary explants of epithelial cells. Minimum Essential Medium (MEM) with Earle’s Balanced Salts is a modification of Eagle’s earlier Basal Medium (BME) which contains a higher concentration of essential nutrients. Studies to determine the nutritional requirements of many cells have been in progress since Eagle's first reports. The major essential nutrients were identified, and work became focussed on the media requirements of individual cell types. Many media designed for these purposes are now available. Among the first of these media, developed initially to study hormonal requirements of cells in culture, was a mixture of DMEM and Ham's F12 medium, known as DMEM-Ham's F12.

Basal Medium Eagle is principally used for monolayer diploid or primary cell cultures. It is the simplest of the basal media with all essential components for growth. BME ideally supports cell lines such as HeLa, L-cells and primary mammalian fibroblasts. BME has also successfully been used for the growth of human transformed cell lines (particularly WI-38 and MRC-5).

The Leibovitz L-15 media were formulated to promote the cell growth in medium not balanced in CO₂. The formulations were developed with the sodium bicarbonate buffer. The Leibovitz L-15 media are buffered by a complement of salts, free base amino acids and galactose so they can be used under conditions of free gaseous exchange with the atmosphere. When properly supplemented, L-15 Medium supports established cell lines, such as HEp-2 and LLC-MK2, as well as primary explants of embryonic and adult human tissue. Many viruses also have been successfully cultivated in this medium.
