Schaeffer and Fulton Spore Stain Solution B, Sigma-Aldrich®
Supplier: SIGMA ALDRICH MICROSCOPY
In Schaeffer-Fulton`s method, a primary stain-malachite green is forced into the spore by steaming the bacterial emulsion. Malachite green (Schaeffer & Fulton`s Spore Stain A: Cat. No. 90903) is water soluble and has a low affinity for cellular material, so vegetative cells may be decolorized with water. Vegetative cells are then counterstained with safarin. The components and the method are also available as kit (Cat. No. 04551).

Tetramethylrhodamine isothiocyanate Isomer R (6-TRITC isomer R), Sigma-Aldrich®
Supplier: SIGMA ALDRICH MICROSCOPY
Tetramethylrhodamine isothiocyanate Isomer R (TRITC) is the R isomer of tetramethylrhodamine isothiocyanate and is also known as 6-tetramethylrhodamine isothiocyanate. It is a strongly fluorescent compound soluble in ethanol, methanol, dimethylformamide, dimethylsulfoxide, and water. TRITC is a moderately-sized aminoxanthene dye carrying carboxylic acid and isothiocyanate substituents on the non-planar pendant phenyl ring. It forms a lipophilic cation in acidic pH and a hydrophilic zwitterion around neutral pH. The isothiocyanate group reacts with nucleophilic substituents (e.g., amino, hydroxyl, thiol) of biomolecules, providing the means of attaching a fluorescent label. TRITC also reacts with water and hydroxide ions and is unstable in aqueous solutions. TRITC is commercially available as a zwitterion or as a chloride.

Formalin 10% histological fixative, neutral buffered
Supplier: SIGMA ALDRICH MICROSCOPY
10% Neutral buffered formalin (NBF) is the most commonly used fixative for light microscopy. 10% NBF is well suited for large throughput laboratories and requires a relatively short period of fixation, but can also be used for the long-term storage of tissue. Formalin fixation is thought to form between the aldehydes and the proteins, creating a gel, thus retaining cellular constituents in their in vivo relationship.

Bromocresol green sodium salt 0.04% in aqueous solution, Sigma-Aldrich®
Supplier: SIGMA ALDRICH MICROSCOPY
Bromocresol green is a sulfonephthalein dye. It is mainly used for measuring serum albumin. In acidic conditions, bromocresol green interacts with albumin, thereby resulting in an increase in absorbance at 630 nm. The change in absorbance is linked with the concentration of albumin. Bromocresol Green is also a pH indicator, changing from yellow (pH 3.8) to blue color (5.4). It is widely used as a colloidal pigment.

Poly-L-lysine hydrobromide
Supplier: SIGMA ALDRICH MICROSCOPY
Poly-L-lysine is a positively charged amino acid polymer with approximately one HBr per lysine residue. The hydrobromide allows the poly-L-lysine to be in a crystalline form soluble in water. A small amount of product may be found in the beta structure because the HBr interferes with hydrogen bonding between amino and either the carboxyl groups or N or O containing moieties.
