Giemsa is a versatile polychromatic stain, which is suitable for staining a diverse range of specimens. It is a buffered thiazine-eosinate solution designed to provide coloration of blood cells. Giemsa’s stain is frequently used for diagnostic purposes in hematology to differentiate the nuclear and cytoplasmic morphology of platelets, RBCs, WBCs, and parasites. It is frequently used in combination with other dye solutions as May-Grunwald’s solution for Pappenheim (MGG) and Wright-Giemsa. The Giemsa stain was adapted to histology due to its unique staining of chromatin, nuclear membranes, and cytoplasmic elements. Giemsa′s staining solution is composed of methylene blue, azure, and eosin.

The tablets are dissolved in 100 ml of the water sample to be tested, which has been buffered at pH 10 with 1 ml of Ammonia/ Ammonium chloride buffer solution, and then titrated with EDTA solution (0,01 M) till the sample turns from red to blue.
Total hardness in mg/l = 10×ml of titrant used.
Control samples of calcium carbonate should be used to calibrate the method.
