Immobilised Protein A and G are designed for binding the constant domains of immunoglobulin (Ig) molecules. This lab activity has two objectives - to deepen the understanding of protein molecules and for students to learn the potential of electrophoresis in protein analysis. Students study the fundamentals of protein structure from their primary structure to the more complex tertiary and quaternary structures, utilising protein electrophoresis. Complex mixtures of protein samples and characterised pure protein samples, in conjunction with electrophoresis, are utilised to study protein structure and the potential of protein electrophoresis. Using non-denaturing and denaturing electrophoresis, students understand the difference between primary, tertiary and quaternary structures, the importance of disulfhydryl bridges in maintaining protein structure and electrophoresis in studying complete proteins and protein subunits.

Pfu DNA Polymerase, derived from the hyperthermophilic archae Pyrococcus furiosus, has superior thermostability and proofreading properties compared to other thermostable polymerases. It has a molecular weight of 90 kDa and can amplify DNA targets up to 2 kb. The elongation velocity is 0,2~0,4 kb/min (70~75 °C). Pfu DNA Polymerase possesses 3' to 5' exonuclease proofreading activity that enables the polymerase to correct nucleotide misincorporation errors. This means that Pfu DNA Polymerase-generated PCR fragments will have fewer errors than Taq-generated PCR inserts. Using Pfu DNA Polymerase results in blunt-ended PCR products, which are ideal for cloning into blunt-ended vectors. Pfu DNA Polymerase is superior for techniques that require high-fidelity DNA synthesis.
