Biodiversity lab activity is designed for determination of protein contents and biomass in diverse biological samples to study how biomass is related to biodiversity in nature. Students collect and catalogue plant leaf samples from a diverse group of locally available plants. This lab activity involves determination of natural weight of each plant sample, grinding a predetermined amount of each sample, and the subsequent extraction of proteins from the samples. Students then learn to determine protein contents of each plant sample and attempt to relate the protein content with biomass. Students in this lab activity are challenged to think, analyse, and seek answers as to why protein biomasses vary for a given natural weight for different plants. Finally, they will relate that finding to the biodiversity of nature.

The stain uses an enhanced Periodic Acid-Schiff (PAS) method for detection of glycoprotein sugars. The supplied agent first oxidises the cis-diol sugar groups to aldehydes. The aldehyde groups react with the sensitive glycoprotein stain solution forming Schiff bonds and producing strong magenta colour bands. RAPIDstain™ can be used after glycoprotein staining to detect non-glycosylated proteins and the use of the stain enhances glycoprotein staining. The glycoprotein stainis highly convenient as all the key reagents required for staining are supplied and a unique positive & negative control is included.

This lab activity is designed to demonstrate the different classes of protein molecules and their classification based on solubility. Students learn fractionation of soluble, insoluble membrane proteins, and cytoskeleton proteins from a tissue sample. The insoluble protein fraction is further fractionated into hydrophilic and hydrophobic membrane proteins. Cell membrane structure and the role of hydrophobic membrane proteins are considered. This lab activity also provides an opportunity to understand characteristics of various classes of detergents and the role of detergents in solubilisation of hydrophobic membrane proteins.

Magenta-gal is used as an alternative to X-gal that produces a red color. Magenta-Gal is hydrolysed by beta-galactosidase to produce a red precipitate. This precipitate is insoluble in alcohol and xylenes, thus making it suitable for immunoblotting and immunocytochemical assays. Bacterial colonies containing active beta-galactosidase produce red colonies when grown on plates containing magenta-gal. Magenta-gal is commonly used in conjunction with IPTG for red/white screening.

SG-Arginine-C™ endopeptidase (Clostripain, from C. histolyticum) specifically hydrolyses the carboxy peptide bond of arginine. SG-Arginine-C™ has been modified chemically by a propriety process to render the enzyme resistant to autolysis and stabilise enzymatic activity. In addition, as a sulfhydryl enzyme, SG-Arginine-C™ is susceptible to inactivation by oxidation and as a result requires reducing agents for protection. The enzyme also requires calcium ion for maximal activity. A special reconstitution buffer is supplied, which contains reducing agents and activators to maintain enzyme activity.
