Elektroforesebuffere opretholder stabile pH-niveauer under adskillelse af DNA, RNA og andre proteiner. Ved at levere ioner, der overfører den påførte strøm, gør de lavladede opløsninger det muligt for molekyler af forskellige størrelser at bevæge sig med forskellige hastigheder til analyse. Som ekstra beskyttelse af proteiner forhindrer de præparative stoffer nedbrydning og sikrer dermed prøvernes levedygtighed og nøjagtige testresultater. Tilgængelige i forskellige molekylvægte kan den bedste elektroforesebuffer vælges ud fra de specifikke makromolekyler, der håndteres.
