Antigen-antistof-tests er baseret på, at der er et specifikt antistof for hvert antigen, og derfor kan hvert antistof bruges til at detektere tilstedeværelsen af det andet. Disse tests udføres normalt på serum, men lejlighedsvis kan prøver af andre kropsvæsker, væv eller afføring anvendes.
