Membrane-permeant form of Rhod-2 that can be loaded into cells via incubation. Because of the relatively low water solubility of the AM ester, the mild detergent Pluronic F-127 is often used as a dispersing agent to facilitate dye loading. Rhod-2-AM itself does not bind Ca2+ , but it is readily hydrolyzed to Rhod-2 by endogenous esterases once it enters cells.

EZ-Vision® 1 kb DNA ladder is a pre-mixture of DNA marker, loading buffer and a proprietary fluorescent DNA visualisation dye that can be applied directly to the gel. This non toxic, non mutagenic dye eliminates the need for ethidium bromide staining. The 1 kb ladder consists of 10 double-stranded bands with molecular weights between 200 and 10 000 bp. The intensity of individual bands varies to facilitate easy identification.

Blue light transilluminators are often used as an alternative to UV transilluminators when users wish to use ‘safe dyes’ instead of ethidium bromide. A blue light transilluminator is a ‘safe’ light source, in that the user is not exposed to harmful UV radiation and samples are free from photonicking. Concanavalin A (Con A) agarose consists of Con A coupled to 6% agarose by the cyanogen bromide method. Con A is a tetrameric metalloprotein lectin isolated from Canavalia ensiformis (jack bean). It is used for the purification of glycoproteins, polysaccharides and glycolipids as it binds molecules containing α-D-mannopyranosyl, α-D-glucopyranosyl and sterically related residues. It has also be used in other application areas including purification of enzyme-antibody conjugates, purification of IgM and separation of membrane vesicles. As stated above, it is a metalloprotein and to maintain its binding characteristics the presence of both Mn²⁺ and Ca²⁺ is essential. Each subunit of Con A utilises one calcium and one manganese ion and these cations can be removed under acidic conditions abolishing the carbohydrate-binding activity.

Ethidium bromide(EtBr) has been used extensively to reduce mitochondrial DNA copy number in proliferating cells and most commonly used in nucleic acid stain for PAGE or agarose gel electrophoresis. The fluorescence of EtBr increases 21-fold upon binding to double-stranded RNA and 25-fold on binding double-stranded DNA so that destaining the background is not necessary with a low stain concentration (10 μg/mL). Ethidium bromide has been used in a number of fluorimetric assays for nucleic acids. It has been shown to bind to single-stranded DNA (although not as strongly) and triple-stranded DNA.4 Because of its ability to bind to DNA, EtBr is an inhibitor of DNA polymerase. A 10mg/mL easy-to-use solution of ethidium bromide in specially filtered, deionized water which is excellent for nucleic acid electrophoresis and purification applications, eliminates the dust hazard associated with powdered ethidium bromide and saves time spent on weighing and mixing. Used in the fluorometric detection of double stranded nucleic acids in gel electrophoresis. Also acts as an RNA polymerase inhibitor, and in separation of high molecular weight DNA's. It confers a deep red stain to DNA by acting as an intercalating agent between the starch bases. Subsequent fluorescence techniques enable the extraction and characterization of DNA bands. Biochem/physiol Actions: Ethidium bromide intercalates double-stranded DNA and RNA and acts as a frameshift mutagen. It can also be used in conjunction with acridine orange to differentiate between viable, apoptotic and necrotic cells.
