This antibody recognises a protein of 25 to 26 kDa, identified as the Bcl-2 alpha oncoprotein. It shows no cross-reaction with Bcl-x or Bax. Expression of Bcl-2 alpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of the protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, Bcl-2 antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express Bcl-2 and the small number in which the neoplastic cells are negative.

This antibody is specific to SUMO1 and shows no cross-reaction with either SUMO2 or SUMO3. The small ubiquitin-related modifier (SUMO) proteins, which include SUMO1, 2 and 3, belong to the ubiquitin-like protein family. Like ubiquitin, the SUMO proteins are synthesised as precursor proteins that undergo processing before conjugation to target proteins. Also, both utilize the E1, E2, and E3 cascade enzymes for conjugation. However, SUMO and ubiquitin differ with respect to targeting. Ubiquitination predominantly targets proteins for degradation, whereas sumoylation targets proteins to a variety of cellular processing, including nuclear transport, transcriptional regulation, apoptosis and protein stability. The unconjugated SUMO1 protein localizes to the nuclear membrane.

CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterised by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45RB is expressed on mature B-lymphocytes and the majority of lymphomas and leukemias of B-cell origin. This antigen is also expressed on the surface of cells of myeloid lineage.

Expression of smooth muscle myosin is developmentally regulated, appearing early in smooth muscle development, and is specific for smooth muscle development. Two isoforms of smooth muscle myosin heavy chain have been identified, designated MHC-1 and MHC-2. The antibody may be useful for the study of breast tumors as the presence of an intact layer of myoepithelial cells is an important feature, which may distinguish benign breast lesions and carcinoma in situ from invasive tumors.

This mAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. About one third of the Ki-1 positive lymphomas lack the leukocyte common antigen (CD45).

Recognises a carbohydrate determinant of Gal 1-3(Fuc 1-4) GlcNAc which is blood group antigen Lewis A. It is present primarily on epithelial cells such as colon and kidneys. In the tumors and dedifferentiated tissues, decrease of Lewis A antigen was observed. Lewis A (type 1 chain) is expressed in colonic epithelial cells and may be useful for detection of gastrointestinal tumors, pancreatic cancer, and colorectal tumors. Blood group related antigens represent a group of carbohydrate determinants carried on both glycolipids and glycoproteins. They are usually mucin-type, and are detected on erythrocytes, certain epithelial cells, and in secretions of certain individuals. Sixteen genetically and biosynthetically distinct but inter-related specificities belong to this group of antigens, including A, B, H, Lewis A, Lewis B, Lewis X, Lewis Y, and precursor type 1 chain antigens.

Recognises a glycoprotein of ~200 kDa, identified as carbonic anhydrase IX (CAIX/CA9/gp200). Its epitope resides in the carbohydrate domain of gp200. It shows no significant cross-reactivity with other carbohydrate determinants, such as the Lewis blood group antigens, epithelial membrane antigen, HMFG, and AB blood group antigens. In normal kidney, CA9 is localized along the brush border of the pars convoluta and pars recta segments of the proximal tubule, as well as focally along the luminal surface of Bowman's capsule adjoining the outgoing proximal tubule. Reportedly, CA9 is expressed by 93% of primary and 84% of metastatic renal cell carcinomas. This mAb may be useful in the investigations of carcinomas of proximal nephrogenic differentiation especially those showing tubular differentiation.

PECAM-1, or platelet endothelial cell adhesion molecule-1, also called CD31, is made up of six extracellular immunoglobulin-like domains, a single transmembrane domain, and a cytoplasmic domain containing serine and tyrosine residues that when phosphorylated, activate the molecules signaling function. It is expressed on platelets, monocytes, neutrophils, bone marrow macrophage and dendritic cells precursors, and at the intracellular junction of endothelial cells. PECAM-1, on the endothelial lining of blood vessels, helps maintain the vascular barrier. In response to inflammatory mediators, the endothelial pull away from each other allowing fluid, inflammatory cells and proteins into the extravascular space. The inflammatory response fades as adhesion of neighboring cells increase. PECAM-1 also plays an important role in angiogenesis, promoting endothelial cell motility, junctional development and capillary morphogenesis.

CD45RA is an isoform of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4+ T-cells, 50% of peripheral CD8+ T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4+ or CD8+ T-cells. It can especially be used to differentiate T-cell lymphomas (CD45RO +ve) from B cell lymphomas (CD45RA +ve).

This mAb recognises a protein of 130 kDa, identified as Ksp-cadherin. Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm. Cadherins have been implicated in embryogenesis, tissue morphogenesis, tissue structure maintenance, cell polarization, neoplastic invasiveness and metastasis, and membrane transport. It is suggested that Ksp-cadherin is a marker for terminal differentiation of the basolateral membranes of renal tubular epithelial cells. Within the kidney, Ksp-Cadherin is found exclusively in the basolateral membrane of renal tubular epithelial cells and collecting duct cells, and not in glomeruli, renal interstitial cells, or blood vessels. Ksp-Cadherin has been suggested to distinguish Chromophobe Renal-Cell Carcinoma from Oncocytoma.

This mAb recognises a protein of 165 kDa, identified as carbamoyl phosphate synthetase 1 (CPS1). This mitochondrial enzyme catalyses synthesis of carbamoyl phosphate from ammonia and bicarbonate. This reaction is the first committed step of the urea cycle, which is important in the removal of excess urea from cells. Deficiency of CPS1 is an autosomal recessive disorder that causes hyperammonemia. CPS1 is a hepatocyte specific protein that localizes to the mitochondria of hepatocytes. It is a sensitive marker for distinguishing hepatocellular carcinomas (HCC) from other metastatic carcinomas as well as cholangio-carcinomas. HCC s occur primarily in the stomach, but they are also found in many other organs. CPS1 may also be a useful marker for intestinal metaplasia. Reportedly, strong expression of CPS1 correlates with smaller tumor size and longer patient survival. Occasionally, CPS1 is also found in gastric carcinomas as well as in a few other non-hepatic tumors.

Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. Estrogen receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of ligand-activated transcription factors. Estrogen receptors, including alpha and beta, contain DNA binding and ligand binding domains and are critically involved in regulating the normal function of reproductive tissues. They are located in the nucleus, though some estrogen receptors associate with the cell surface membrane and can be rapidly activated by exposure of cells to estrogen. ER alpha and beta are differentially activated by various ligands. Ligand interaction triggers a cascade of events, including dissociation from heat shock proteins, recepter dimerization, phosphorylation and the association of the hormone activated receptor with specific regulatory elements in target genes. Evidence suggests that ER alpha and beta may be regulated by distinct mechanisms even though they share many functional characteristics.
