These drugs bind to CRBN, leading to the degradation of transcription factors Ikaros and Aiolos, which are essential for myeloma cell survival. Additionally, novel cereblon E3 ligase modulators (CELMoDs) are being developed to overcome resistance to IMiDs, offering new therapeutic options for patients with CRBN mutation.
