DY677 CEP (DyLight 677 phosphoramidite) is an excellent replacement to the commonly used Cy5.5 phosphoramidite.

DY677 CEP appears to be more stable than Cy5.5 phosphoramidite. As Cy5.5 phosphoramidite, the deprotection of oligos containing DY677 CEP may be carried out with ammonium hydroxide at room temperature, regardless of the base protecting groups on the monomers used.

It is always prudent to use monomers with base labile protecting groups to limit the exposure time to 2 hours or less at 65 °C during deprotection.
