L'uso degli enzimi batterici caratteristici offre la possibilità di una rapida identificazione. in quanto gli enzimi mostrano una notevole attività indipendentemente dalla condizioni di crescita. Sotto il marchio Fluorocult®, Merck offre un'ampia gamma di terreni di coltura solidi e liquidi che consentono il rilevamento in fluorescenza anche del batterio E. coli. I terreni di coltura Fluorocult® hanno la medesima composizione dei terreni standard con in più il MUG un substrato fluorogeno che viene metabolizzato da enzimi batteri specifici. I batteri diventano in tal modo visibili in fluorescenza. Alcuni terreni contengono anche substrati con triptofano per un'ulteriore conferma della presenza del batterio E. coli tramite eventuale reazione dell'indolo. Il metodo Fluorocult® può produrre un risultato certo in sole 24-48 ore. In caso di terreno cromogeno (contenente un substrato che, una volta metabolizzato da enzimi specifici, rende i batteri identificabili grazie al cambiamento di colore), è possibile effettuare un'identificazione diretta grazie alla colorazione caratteristica delle colonie sullo stesso terreno di coltura senza l'uso di ulteriori additivi. È possibile ottenere una differenziazione delle singole colonie positive anche in presenza di elevati conteggi microbici.
