CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 β chain (CD8 α/β) or as a homodimer (CD8 α/α). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.

Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

Recognizes a protein of 205 kDa-220 kDa, identified as CD45RA. CD45RA is isoforms of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4 T-cells, 50% of peripheral CD8 T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4 or CD8 T-cells. It can especially be used to differentiate T-cell lymphomas (CD45RO ve) from B cell lymphomas (CD45RA ve).

By immunohistochemistry, this antibody specifically recognizes a protein in melanocytes and melanomas. Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD28 is also expressed on human fetal NK cells and some NK cell lines, whereas on murine NK cells the CD28 expression is much broader.

This antibody recognizes a 24-27 kDa estrogen-regulated protein, identified as heat shock protein 27 (hsp27). Hsp27 was recently found to be identical to the estrogen-induced p29 and 24K protein. About 50% of breast carcinomas are positive for hsp27 especially those that are also positive for estrogen and/or progesterone receptor. HSP27 has also been implicated in drug resistance in cancer cells.

Reacts with five invariable CD3 chains (designated as and ) with molecular weight ranging from 16-28 kDa. CD3 is expressed, typically at high levels, on peripheral T cells and majority of T cell neoplasms. Thymocytes express CD3 at different level on the cell surface in the course of differentiation and, in cortical thymus, CD3 is predominantly Intracytoplasmic. The CD3 complex is closely associated at the lymphocyte cell surface with T cell antigen receptor (TCR) and is involved in transducing antigen-recognition signals into cytoplasm of T cells and in regulating the cell surface expression of the TCR complex.

This antibody recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43. 70-90% of T-cell lymphomas and from 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.

Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO). This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. This antibody recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Grp94 has been briefly studied as a prognostic factor in breast cancer.

CD2 interacts through its amino-terminal domain with the extracellular domain of CD58 (also designated CD2 ligand) to mediate cell adhesion. CD2/CD58 binding can enhance antigen-specific T cell activation. CD2 is a transmembrane glycoprotein that is expressed on peripheral blood T lymphocytes, NK cells and thymocytes. CD58 is a heavily glycosylated protein with a broad tissue distribution in hematopoietic and other cells, including endothelium. Interaction between CD2 and its counter receptor LFA3 (CD58) on opposing cells optimizes immune system recognition, thereby facilitating communication between helper T lymphocytes and antigen-presenting cells, as well as between cytolytic effectors and target cells.
