L'E.Z.N.A.® Plasmid Midi Kit rappresenta un metodo affidabile e di recente sviluppo per isolare fino a 200 µg di DNA plasmidico in meno di 60 minuti. Il kit combina una tecnologia a membrana HiBind® con la costanza verificata nel tempo della lisi SDS alcalina di cellule batteriche per fornire DNA plasmidico di alta qualità. Le colonne HiBind® DNA midi prive di DNasi/RNasi funzionano eliminando le lunghe estrazioni con fenolo/cloroformio e le precipitazioni con alcol. La purificazione del plasmide con l'E.Z.N.A.® Plasmid Midi Kit segue una semplice procedura di legame-lavaggio-eluizione, rendendo quindi possibile la processazione contemporanea di campioni multipli. Sebbene le rese varino in base al numero di copie del plasmide, al ceppo E.coli e alle condizioni di crescita, una coltura notturna di 50 ml in terreno LB produce generalmente 200 µg di DNA plasmidico con alto numero di copie. Quando si lavora con plasmidi a basso numero di copie, il volume della coltura iniziale può essere aumentato di massimo 100 ml. The Mag-Bind® Plant DNA DS 96 Kit is designed for difficult samples (DS) which are hard to lyse or contain high amounts of polysaccharides and polyphenols. The system uses a CTAB-based lysis buffer which does not require organic solvents. The proprietary binding system prevents inhibitors from binding to the magnetic beads.
