Resistendo alle variazioni di pH, i tamponi proteggono i campioni quando vengono aggiunti acidi o basi. Disponibili in polvere e in forma liquida, tracciano efficacemente le proteine durante l'isolamento e la separazione tramite elettroforesi. I tamponi salati possono anche essere utilizzati per calibrare e standardizzare i pHmetri per ottenere letture accurate. Le sostanze bloccanti impediscono l'adesione degli anticorpi ai siti liberi dalla membrana, che produrrebbero un background interferente.
