Nuclease S1 is an endonuclease that is active against single-stranded DNA and RNA molecules. Although its primary substrate is single-stranded, it can also introduce single-stranded breaks in double-stranded DNA-RNA or DNA-RNA hybrids. Nuclease S1 is used in the laboratory as a reagent in nuclease protection assays. It is used in removing single stranded tails from DNA molecules to create blunt ended molecules and opening hairpin loops generated during synthesis of double stranded cDNA.

Protease S.aureus V8 (endoproteinase-glu-C) specifically cleaves peptide bonds on the COOH-terminal side of either aspartic or glutamic acids. In the presence of ammonium, the enzyme specificity is limited to glutamic sites. It has a molecular weight of 27000 daltons and optimum pH's of 4,0 and 7,8 with hemoglobin as the substrate. Enzyme activity is determined by the casein digestion assay.

For DNase digestion during RNA purification. The RNase-free DNase set provides a rapid and efficient on-column digestion of DNA during RNA purification from a wide variety of sample sources using E.Z.N.A.® RNA kits. The DNases can be efficiently removed in subsequent steps. Generally, DNase digestion is not required for RNA purified with E.Z.N.A.® RNA kits since the silica-membrance, spin-column technology efficiently removes the majority of the DNA without DNase treatment. However, complete DNA removal may be necessary for certain RNA applications that are sensitive to very small amounts of DNA.

Two types of 15-lipoxygenase (15-LO) have been discovered and characterised, both of which metabolise arachidonic acid (AA) to produce 15(S)-hydroxyeicosatetraenoic acid (15(S)-HETE). 15-LO-1 oxygenates AA at both C-15 and C-12, whereas 15-LO-2 exclusively oxygenates C-15 of AA. Human 15-LO-2 has a molecular mass of approximately 76 kDa and exhibits approximately 40% identity to the reticulocyte 15-LO-1.2,3 expression of 15-LO-2 appears to be restricted to prostate, lung, skin, and cornea and may play a role in the normal development of these tissues.
