Gram’s safranin solution, also known as Safranin T or Basic Red 2, is a ready-to-use staining solution that comprises 10% safranin in ethanol. Safranin is a basic, cationic, azine dye containing a planar, diaminophenazine chromophore, with a phenyl substituent attached to a ring nitrogen. The trimethyl cationic form of safranine is lipophilic, whereas the dimethyl forms are weakly hydrophilic. It is most notably used in bacteriology as a counterstain in Gram staining to differentiate between Gram-positive and Gram-negative bacteria.

Alcian Blue solution has been used in the histological staining of acidic glycosaminoglycans (GAGs; mucopolysaccharides) that are carboxylated or sulfated. Sulfated glycosaminoglycans appear to be the preferred substrate for alcian blue at pH 2.5. Alcian blue has been used to stain hyaluronan (HA), a nonsulfated GAG, but the intensity of the staining of HA is affected by the method of tissue fixation used. Alcian blue staining of GAGs in tissue may be modified through the addition of an electrolyte such as magnesium chloride.

The stain uses an enhanced Periodic Acid-Schiff (PAS) method for detection of glycoprotein sugars. The supplied agent first oxidises the cis-diol sugar groups to aldehydes. The aldehyde groups react with the sensitive glycoprotein stain solution forming Schiff bonds and producing strong magenta colour bands. RAPIDstain™ can be used after glycoprotein staining to detect non-glycosylated proteins and the use of the stain enhances glycoprotein staining. The glycoprotein stainis highly convenient as all the key reagents required for staining are supplied and a unique positive & negative control is included.

Oil red O is a lysochrome (fat-soluble dye) diazo dye used for staining of neutral triglycerides and lipids on frozen sections and some lipoproteins on paraffin sections. In histology, a supersaturated solution of oil red O in isopropanol may be used to stain fat in tissue.

Wright′s Stain solution is commercially available and is generally used in hematology. Its preparation involves heating methylene blue with sodium bicarbonate and combining the mixture with eosin. It is alkaline, hence, its concentration is continuously changing due to the progressive oxidizing action of the alkali. Therefore, Giemsa′s stain is added to preserve the azurophilic properties of polychrome dyes.

Brilliant Blue G is a methanol based stain which is especially designed for staining protein in polyacrylamide gels, but will also stain protein in agarose gels (IEF).
