La coloration des bactéries de Gram constitue l'un des principaux tests en microbiologie et souvent l'un des premiers tests pratiqués dans l'identification des bactéries. La méthode de coloration de Gram fait appel aux propriétés des parois des cellules bactériennes et au colorant cristal violet. Les parois des microorganismes à Gram positif ont une plus grande quantité de peptidoglycanes et une teneur lipidique inférieure aux bactéries à Gram négatif, ce qui leur permet de retenir le colorant de Gram. Streptavidin Resin is designed for the single step small and large scale affinity purification of proteins and antibodies with a biotin tag. Biotin, a 244 Da vitamin (Vitamin H) molecule, exhibits an extraordinary binding affinity for avidin (Ka=10¹⁵ M⁻¹) and streptavidin (Ka=10¹⁵ M⁻¹). Biotin and (strept)avidin interaction is rapid and once the bond is established it can survive up to 3M guanidinehydrochloride and extremes of pH. Biotin-avidin bonds can only be reversed by denaturing the avidin protein molecule with 8M guanidine-hydrochloride at pH 1,5 or by autoclaving. The biotinylated molecules are efficiently probed with avidin or streptavidin conjugated to reporter molecules, such as peroxidases or phosphatases.

Ils assimilent la précipitation des protéines due au pH, à une grande concentration de sel et en présence de solvants organiques, et la dénaturation des protéines résultant d'une température élevée. De plus, le kit démontre dans quelle mesure les non-protéines, comme les détergents, modifient considérablement les propriétés physiques des molécules de protéines, ce qui explique l'importance de ces détergents dans la solubilisation des protéines. It uses chlorophenol red-β-D-galactopyranoside (CPRG), which is up to 10 times more sensitive than the classic O-nitrophenyl-β-D-galactopyranoside (ONPG) β-Galactosidase assays. This increased sensitivity makes it simpler to measure β-galactosidase activity in cells that are difficult to transfect or have low β-galactosidase activity expression. β-Galactosidase activity is measured using a spectrophotometer or a microplate reader to determine the amount of substrate converted at 570 to 595 nm.
