Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

This mAb recognises a protein of about 50 kDa which is identified as gamma-Enolase/Neuron Specific Enolase/Enolase 2. Three isoenzymes of enolases are identified, alpha, beta and gamma. Alpha-isoform is expressed in most tissues, whereas beta-form is expressed predominantly in muscle tissue and gamma-enolase is found only in nervous tissue. These isoforms exist as both homodimers and heterodimers, and they play a role in converting phosphoglyceric acid to phosphenolpyruvic acid in the glycolytic pathway. NSE is a useful marker to identify peripheral nerves and tumors of neuro-endocrine origins, such as pheochromocytomas. It it be usually employed in combination with other markers such as Synaptophysin, Chromogranin A, and Neurofilament.

Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm. Cadherins have been implicated in embryogenesis, tissue morphogenesis, tissue structure maintenance, cell polarization, neoplastic invasiveness and metastasis, and membrane transport. It is suggested that Ksp-cadherin is a marker for terminal differentiation of the basolateral membranes of renal tubular epithelial cells. Within the kidney, Ksp-Cadherin is found exclusively in the basolateral membrane of renal tubular epithelial cells and collecting duct cells, and not in glomeruli, renal interstitial cells, or blood vessels. Ksp-Cadherin has been suggested to distinguish Chromophobe Renal-Cell Carcinoma from Oncocytoma.

Reacts with a 70 kDa membrane-bound isozyme (Regan and Nagao type) of placental Alkaline Phosphatase (PLAP) occurring in the placenta during the 3rd trimester of gestation. Anti-PLAP reacts with germ cell tumors and can discriminate between these and other neoplasms. Somatic neoplasms e.g. breast, gastrointestinal, prostatic, and urinary cancers may also immunoreact with antibodies to PLAP. Anti-PLAP positivity in conjunction with anti-keratin negativity favors seminoma over carcinoma. Germ cell tumors are usually anti-keratin positive, but they regularly fail to stain with anti-EMA, whereas most carcinomas stain with anti-EMA. Anti-PLAP has been useful in the diagnosis of gestational trophoblastic disease.

Expression of TMEM16A (DOG1) protein is elevated in the gastrointestinal stromal tumors (GIST’s), c-kit signaling-driven mesenchymal tumors of the GI tract. It is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG11 has been reported in 97,8 percent of scorable GIST’s, including all c-kit negative GIST’s. Overexpression has been suggested to aid in the identification of GISTs, including Platelet-Derived Growth Factor Receptor Alpha (PDGFR alpha) mutants that fail to express c-kit antigen. The overall sensitivity of TMEM16A and c-kit in GIST’s is nearly identical: 94,4% vs. 94,7%.

This mAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

PTH/Parathyroid hormone is a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood Ca2+ levels by dissolving the salts in bone and preventing their renal excretion. It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterised by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.

Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognises two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.

Recognises a protein of 40 kDa, identified as cytokeratin-19 (CK19), which is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

This antibody recognises a 53 kDa protein, identified as the Ornithine Decarboxylase. ODC-1 is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC-1 mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. Activity in colorectal carcinomas is greater than those in adenomas and normal mucosa.

Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. SCUBE3 Antibody: SCUBE3 is a member of a family of secreted glycoproteins that contain N-terminal EGF-like repeats and C-terminal cysteine-rich motifs and CUB domain and is highly expressed in primary osteoblasts and bones, and to a lesser extent in heart (1,2). Other studies have shown that overexpression of SCUBE3 in mice induced cardiac hypertrophy, suggesting that it may also play a role in the regulation of cardiac growth. SCUBE3 has been shown to be an endogenous TGF-beta receptor ligand (3,4) and is thought to promote lung cancer cell mobility and invasiveness. In lung cancer cells, the secreted SCUBE3 protein was cleaved by MMP2 and MMP9, allowing the activation of the TGF-beta receptor, the increase of Smad2/3 transcriptional activity and the upregulation of expression of proteins such as TGF-beta1, VEGF, Snail, and Slug.
