CEA-related cell adhesion molecules (CEACAM) belong to the carcinoembryonic antigen (CEA) family. It consists of seven CEACAM (CEACAM1, CEACAM3-CEACAM8) and 11 pregnancy-specific glycoprotein (PSG1-PSG11) members. The CEA family proteins belong to the immunoglobulin (Ig) superfamily and are composed of one Ig variable-like (IgV) and a varying number (0-6) of Ig constant-like (IgC) domains. CEACAM molecules are membrane-bound either via a transmembrane domain or a glycosyl phosphatidyl inositol (GPI) anchor. CEACAM molecules are differentially expressed in epithelial cells or in leukocytes. Overexpression of CEA/CEACAM5 in tumors of epithelial origin is the basis of its wide-spread use as a tumor marker. The function of CEACAM family members varies widely: they function as cell adhesion molecules, tumor suppressors, regulators of lymphocytes and dendritic cell activation, receptors of Neisseria species and other bacteria.

BAP31 (B cell receptor-associated protein 31) is an integral protein of the ER membrane that forms a large hetero-oligomeric complex with the related BAP29. The cytosolic domain of BAP31 contains two identical caspase recognition sites that are preferentially cleaved by initiator caspases, including caspase-8. After activation of cell surface death receptors, human BAP31 is cleaved into a membrane-embedded fragment called p20, which induces apoptosis when expressed ectopically. The p20 fragment can mediate Ca2+-dependent apoptotic cross-talk between the ER and mitochondria, stimulating cytochrome c release. In addition to its role in apoptosis, BAP31 is a component of the ER quality control compartment and functions as a cargo receptor for ER export of transmembrane proteins such as cellubrevin, MHC class I molecules, and cytochrome P450 2C2.

A predominant cleaved form of pro-glucagon found in the pancreas and central nervous system. It has been reported that human pancreatic endocrine tumours contain molecular forms of GLP-1 which resemble those expressed in the small intestine, rather than in their tissue of origin. A number of E2s in Saccharomyces cerevisiae and their homologues have been identified. One such family of E2s is the UBC4/5, characterised as essential for the degradation of short-lived, regulatory and abnormal proteins. Protein levels of S. cerevisiae UbC4/5 are up-regulated in response to stress, and their loss results in severe effects on cellular functions. A human gene product that is 79% identical to S. cerevisiae UBC4/5 in amino-acid sequence was identified as UbcH5a. In addition, two other human members of this highly conserved E2 class were also cloned and designated as UbcH5b and UbcH5c, having 88% and 89% identity to UbcH5a, respectively. Members of the UbcH5a/b/c are the most active class of E2s in cell extracts. The importance of this enzyme class is underscored by the critical role of UBC4/5 in S. cerevisiae. UbcH5a stimulates the conjugation of ubiquitin to the tumour-repressor p53 in the presence of E6-AP and E6. Moreover, UbcH5 family is implicated in c-fos recognition, the modulation of which is controlled by the ubiquitin-proteasome pathway. UbcH5b and UbcH5c are associated with the signal-induced conjugation and subsequent degradation of IkBalpha in the presence of the SCF complexes. UbcH5c also catalyses the ubiquitination leading to the processing of p105 precursor to form p50, a subunit of the heterodimeric transcription factor NF-kB. The range and diversity of substrates and E3s with which this class of E2 enzymes interact, suggest their complex roles in cellular functions require to be studied further.
