Schneider’s Insect Medium was developed to support the growth of excised imaginal discs from the fruit fly, Drosophila melanogaster. When supplemented with 5 to 20% heat-inactivated foetal bovine serum Schneider's mediuam has been found to support the rapid growth of both primary and established cultures of cells derived from Drosophila spp. and several other dipterans.

The major essential nutrients were identified, and work became focussed on the media requirements of individual cell types. Many media designed for these purposes are now available. Among the first of these media, developed initially to study hormonal requirements of cells in culture, was a mixture of DMEM and Ham's F12 medium, known as DMEM-Ham's F12.

These irrigating buffers and salt solutions are sterile, physiological balanced solutions intended for use in the maintenance of mammalian cells, where a chemically defined, balanced salt solution provides an environment that will maintain the structural and physiological integrity of cells in vitro. Applications include the intermediate steps for cultivation of cells, like washing, centrifugation, suspending and counting, as well as for many analytical methods or biochemical treatments. Principally these solutions should be used to keep the cells in suspension only for minutes or a few hours maximum. These buffers and solutions are not cell culture media.

In 1976, Guilbert and Iscove demonstrated that precursor cells of erythrocytes and macrophages could be cultured in a reduced-serum medium supplemented with albumin, transferrin, lecithin, and selenium. Iscove's medium is a modification of Dulbecco's Modified Eagle's Medium (DME) containing selenium, additional amino acids and vitamins, sodium pyruvate, HEPES buffer, and potassium nitrate instead of ferric nitrate. Further studies have demonstrated that Iscove's medium will support murine B lymphocytes, haemopoietic tissue from bone marrow, B-cells stimulated with lipopolysaccharide, T-lymphocytes, and a variety of hybrid cells.

Newborn Calf Serum (NBCS) originates from 3 to 10 day old calves. It is a high quality, cost effective replacement for Foetal Bovine Serum (FBS) and contains a somewhat higher proportion of proteins and immunoglobulin, but fewer growth factors. With proper cell adaptation, it has been shown to perform very well in cell culture applications where routine cell growth and cell culture maintenance is the objective.
