COL6A2 is one of the three alpha chains of type VI collagen, a beaded filament collagen found in most connective tissues. The protein contains several domains similar to von Willebrand Factor type A domains. These domains have been shown to bind extracellular matrix proteins, an interaction that explains the importance of this collagen in organizing matrix components. Jacalin is an alpha-D-galactose binding lectin extracted from jack-fruit seeds (Artocarpus integrifolia). This lectin is a glycoprotein (approx. 40 kDa) composed of four identical subunits. Jacalin binds to human IgA1 (also called serum or secretory IgA) with high specificity (i.e., it does not bind IgG or IgM). Thus, Jacalin that has been immobilised on crosslinked beaded agarose is useful for purifying human IgA from serum or colostrum samples.

chromatography using antigen coupled to agarose beads. F(ab')2 fragments were generated using a pepsin digestion. Fc fragments and whole IgG molecules have been removed. Fragments were conjugated to DyLight 649. Antibody concentration was determined by extinction coefficient: absorbance at 280 nm of 1,4 equals 1,0 mg of IgG. By immunoelectrophoresis and ELISA this antibody reacts specifically with human F(ab')2. No antibody was detected against nonimmunoglobulin serum proteins. Less than 1% cross reactivity to mouse and rat IgG was detected. This antibody may cross react with F(ab')2 from other species.
